摘 要:目的:分析鉴定添加红曲霉菌发酵前后乌药叶特征成分的变化。方法:首先通过固态发酵制得发酵后乌药叶,采用超高效液相色谱-串联四极杆飞行时间质谱(UPLC-Q-TOF-MS)对发酵前后的乌药叶提取物进行定性分析。UPLC-Q-TOF-MS采用Agilent Eclipse Plus-C18 RRHD色谱柱(2.1 mm × 100 mm,1.8μm),以乙腈-0.1%甲酸水为流动相进行梯度洗脱。流速0.4 mL•min^(-1),柱温40℃,检测波长254 nm,进样量2μL。质谱分析采用电喷雾正离子模式采集。质谱分析采用电喷雾正离子全扫描模式采集,扫描范围为m/z 100-1500Da。结论:本文通过UPLCQ-TOF-MS对乌药叶红曲霉添加发酵前后样品的提取物的化学成分进行定性分析与比较。鉴定出31种主要成分,显著降低乌药叶中原有的黄酮及酚酸类物质,新增了10多种红曲霉菌发酵物中特有的活性成分,主要为莫纳可林类物质。该方法快速、可靠、高效,能较全面地反映红曲霉发酵的乌药叶的化学成分变化,可为深入开发红曲乌药叶产品提供基础依据。Objective:To analyze and identify The changes of characteristic components of R Lindera aggregate leaves be⁃fore and after fermentation with Monascus.Methods:The leaves were prepared by solid state fermentation,and the extracts were qualitatively analyzed by ultra-high performance liquid chromatography-tandem quadrupole time-of-flight mass spec⁃trometry(UPLC-Q-TOF-MS).Agilent Eclipse Plus-C18 RRHD Column(2.1 mm × 100 mm,1.8μm)was used for gradi⁃ent elution with acetonitrile-0.1%formic acid water as the mobile phase.Mass spectrometry analysis was acquired in elec⁃trospray positive ion full scan mode with a scanning range of m/z 100-1500Da.Conclusions:In this paper,the chemical constituents of the extracts of Monascus folium before and after fermentation were qualitatively analyzed and compared by UPLC-Q-TOF-MS.The 31 main components were identified,which significantly reduced the original flavonoid and pheno⁃lic acid substances in the leaves,and added more than 10 kinds of active ingredients in monacolin fermentation,mainly mo⁃nacolin substances.The method is rapid,reliable and efficient,and can reflect the chemical composition changes of Monascus Lindera aggregate leaves,which can provide a basis for further development of Lindera aggregate leaves.
