摘 要:为了研究绞股蓝珍珠粉胶囊对小鼠睡眠的改善作用,本试验采用直接睡眠试验、延长戊巴比妥钠睡眠时间试验、戊巴比妥钠阈下剂量催眠试验和巴比妥钠睡眠潜伏期试验评价绞股蓝珍珠粉胶囊低、中、高剂量组[120、240和480 mg/(kg·bw)]、绞股蓝水提物低、中、高剂量组[112、224和448 mg/(kg·bw)]和珍珠粉低、中、高剂量组[8、16和32 mg/(kg·bw)]对小鼠睡眠的改善作用,并对小鼠脑组织中的5-羟色胺(5-HT)、γ-氨基丁酸(GABA)和多巴胺(DA)含量进行测定。结果显示,各组的直接睡眠试验结果皆为阴性;与空白对照组相比,绞股蓝珍珠粉胶囊各剂量组小鼠戊巴比妥钠睡眠时间均显著延长(P<0.01或P<0.001);戊巴比妥钠阈下催眠剂量入睡小鼠只数显著增加(P<0.05或P<0.01);且小鼠的睡眠潜伏期显著缩短(P<0.01或P<0.001)。另外,绞股蓝珍珠粉胶囊各剂量组小鼠脑组织中5-HT和GABA含量不同程度增加(P<0.05或P<0.01或P<0.001),DA含量不同程度降低(P<0.05或P<0.01)。结果表明,绞股蓝珍珠粉胶囊可能通过调节小鼠脑组织内神经递质达到镇静催眠作用。In this study,the effects of Gynostemma pentaphyllum pearl powder capsules on sleep improvement in mice were investigated.The low-,medium-,and high-dose groups of Gynostemma pentaphyllum pearl powder capsules[120,240,and 480 mg/(kg·bw)],Gynostemma pentaphyllum aqueous extract[112,224,and 448 mg/(kg·bw)],and pearl powder[8,16,and 32 mg/(kg·bw)]were evaluated using direct sleep tests,pentobarbital sodium sleep time extension tests,subthreshold dose-induced sleep tests with pentobarbital sodium,and barbital sodium sleep latency tests.The contents of 5-hydroxytryptamine(5-HT),γ-aminobutyric acid(GABA),and dopamine(DA)in mouse brain tissues were also determined.The results showed that all groups had negative results in the direct sleep tests.Compared with the blank control group,the pentobarbital sodium sleep time of mice was significantly prolonged(P<0.01 or P<0.001),the number of mice falling asleep under the sub-threshold dose of pentobarbital sodium significantly increased(P<0.05 or P<0.01),and the sleep latency of mice was significantly shortened(P<0.01 or P<0.001)in all dose groups of Gynostemma pentaphyllum pearl powder capsules.Furthermore,the contents of 5-HT and GABA in the brain tissues of mice in all doses of Gynostemma pentaphyllum pearl powder capsules increased to varying degrees(P<0.05 or P<0.01 or P<0.001),while DA contents decreased to varying degrees(P<0.05 or P<0.01).These results indicate that Gynostemma pentaphyllum pearl powder capsules may achieve a sedative and hypnotic effect by regulating neurotransmitters in the brain tissues of mice.
