摘 要:本试验旨在探究牛油果油对高脂饲粮诱导的肥胖小鼠体内胆固醇代谢的调节作用及其机制。用水代法提取牛油果油,采用气相色谱-质谱联用法检测其脂肪酸组成;采用60%高脂饲粮喂养C57BL/6J雄性小鼠构建肥胖模型,造模成功后,以325、650和975 mg/(kg·d)剂量的牛油果油分组灌胃4周。灌胃结束后,测定肥胖小鼠的体重和血清生化指标,对肝脏和脂肪组织进行组织病理学观察,并检测肝脏胆固醇代谢相关基因的表达情况。结果显示:牛油果油中的主要脂肪酸成分为油酸、棕榈油酸、亚油酸、棕榈酸和13-十八碳烯酸,其中油酸占比为55.56%。325、650和975 mg/(kg·d)牛油果油干预可显著降低肥胖小鼠体重增加量(P<0.001)和肝脏系数(P<0.05),使肥胖小鼠血清谷丙转氨酶(ALT)活性显著降低(P<0.001),血清高密度脂蛋白胆固醇(HDL-C)含量显著增加(P<0.05),并改善肥胖小鼠肝脏和脂肪组织脂质沉积;此外,650和975 mg/(kg·d)牛油果油干预还可使肥胖小鼠血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)含量和谷草转氨酶(AST)活性显著降低(P<0.01)。在胆固醇代谢相关基因表达方面,325、650和975 mg/(kg·d)牛油果油干预使肥胖小鼠肝脏中FXR的mRNA相对表达量显著下调(P<0.05),肝脏中ATP结合盒转运体G5(ABCG5)、ATP结合盒转运体G8(ABCG8)和胆固醇7α-羟化酶(CYP7A1)的mRNA相对表达量显著上调(P<0.05);650和975 mg/(kg·d)牛油果油干预显著上调了肥胖小鼠肝脏中肝X受体(LXR)、腺苷酸活化蛋白激酶α(AMPKα)的mRNA相对表达量(P<0.05),显著下调了肝脏中固醇调控元件结合蛋白2(SREBP2)的mRNA相对表达量(P<0.05);325和650 mg/(kg·d)牛油果油干预显著下调了肥胖小鼠肝脏中ATP结合盒转运体G1(ABCG1)的mRNA相对表达量(P<0.05);975 mg/(kg·d)牛油果油干预显著上调了肥胖小鼠肝脏中胆固醇27α-羟化酶(CYP27A1)的mRNA相对表达量(P<0.05)。由此可见,牛油果油能调节由�The aim of this study was to explore the regulatory effect of avocado oil on cholesterol metabolism and its mechanism in obese mice induced by high-fat diet.Avocado oil was extracted by water substitution method,and its main fatty acids were determined by gas chromatography-mass spectrometry(GC-MS)method.The obesity model was established by feeding C57BL/6J mice with 60% high-fat diet.After the model was successfully established,the obese mice were fed with avocado oil at the doses of 325,650 and 975 mg/(kg·d)for 4 weeks,respectively.After the gavage,the body weight and serum biochemical indexes were measured,the histopathological observations of the liver and adipose tissue were performed,and the expression of genes related to cholesterol metabolism in liver was detected.The results showed as follows:The main fatty acids in the avocado oil were oleic acid,palmitic acid,linoleic acid,palmitic acid and 13-octadecenoic acid,of which oleic acid accounted for 55.56%.Intervention by 325,650 and 975 mg/(kg·d)avocado oil could significantly decrease the body weight gain(P<0.001),liver coefficient(P<0.05)and serum alanine aminotransferase(ALT)(P<0.001),significantly increase serum high-density lipoprotein cholesterol(LDL-C)content(P<0.05),and aspartate aminotransferase(AST)activity in obese mice(P<0.05),and improve the lipid deposition in liver and adipose tissue of obese mice;moreover,intervention by 650 and 975 mg/(kg·d)avocado oil could significantly decrease serum total cholesterol(TC),triglyceride(TG)contents and aspartate aminotransferase(AST)activity of obese mice(P<0.01).In terms of gene expression related to cholesterol metabolism,intervention by 325,650 and 975 mg/(kg·d)avocado oil could significantly down-regulate the mRNA relative expression level of Farnesoid X receptor(FXR)(P<0.05),and significantly up-regulate the mRNA relative expression levels of ATP-binding cassette G member 5(ABCG5),ATP-binding cassette G member 8(ABCG8)and cholesterol 7 alpha hydroxylase(CYP7A1)in liver of obese mice(P<0.05);inter
