摘  要:试验以女贞子为原料,在单因素试验基础上,采用响应面法优化女贞子总黄酮提取工艺,并研究女贞子总黄酮提取液的体外抗氧化活性。结果表明,女贞子总黄酮最优提取工艺参数为乙醇浓度80%、纤维素酶浓度0.3 g/L、超声温度50℃。在此条件下重复测定3次,女贞子总黄酮提取得率的平均值为4.93%,与模型预测值4.98%高度相符,表明该模型可靠,可用于女贞子中总黄酮的提取。女贞子总黄酮提取液体外抗氧化试验表明,总黄酮提取液对1, 1-二苯基-2-三硝基苯肼(DPPH)自由基清除的半数抑制浓度(IC_(50))值为186.6 mg/L,对2, 2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS~+)自由基清除的IC_(50)值为166.8 mg/L,表明女贞子总黄酮提取液具有较强的抗氧化活性。研究表明,女贞子总黄酮可作为天然绿色抗氧化剂在动物生产中应用。On the basis of single factor test,response surface method was used to optimize the extraction process of Fructus ligustris L.,and the in vitro antioxidant activity of total flavonoids from Fructus ligustris L.was studied.The results showed that the optimal process parameters were ethanol concentration of 80%,cellulase concentration of 0.3 g/L and ultrasonic temperature of _(50)℃.Under these conditions,the average extraction rate of total flavonoids from Fructus ligustris L.was 4.93%,which was highly consistent with the predicted value of 4.98%,indicating that the model was reliable and could be used for the extraction of total flavonoids from Fructus ligustris L..The results of antioxidant test showed that the IC_(50) value of total flavonoids extract was 186.6 mg/L for DPPH free radical scavenging and 166.8 mg/L for ABTS+free radical scavenging,indicating that total flavonoids extract had strong antioxidant activity.The study indicates that total flavonoids of Fructus ligustris L.can be used as a natural green antioxidant in animal production.
