摘  要:以油茶果壳为原料,对其多糖的提取纯化工艺及抗氧化活性进行初步研究,为油茶果壳的实际开发利用提供理论依据。以多糖得率和含量为指标,分别比较甲醇、乙醇和丙酮等不同溶剂及50%,60%,70%,80%和90%等不同体积分数梯度的乙醇水溶液对多糖的纯化效果。采用5%,10%,15%,20%和25%等不同比例(活性炭质量/醇沉粗多糖质量)活性炭对油茶果壳多糖进行初步脱色纯化并筛选出最佳的纯化工艺。以DPPH自由基清除能力、ABTS+自由基清除能力及Fe^(3+)还原能力作为指标,对油茶果壳纯化后的多糖进行体外抗氧化活性的测定。结果表明:在80%乙醇水溶液纯化工艺下,油茶果壳多糖含量为34.93%,得率为39.55%,对ABTS+自由基和DPPH自由基的IC50值分别为45.08μg/mL和68.80μg/mL,多糖浓度2 mg/mL时铁离子还原能力最强;在80%乙醇结合10%活性炭脱色的纯化工艺下,油茶果壳多糖含量为37.10%,得率为16.31%,对ABTS+自由基和DPPH自由基的IC50值分别为57.80μg/mL和38.61μg/m L,多糖浓度2 mg/mL时铁离子还原能力最强。研究结果表明,纯化后的油茶果壳多糖具有较强的抗氧化活性,可作为一种天然抗氧化剂应用在食品和医药行业。To study the extraction and purifcation technology and antioxidant activity of Polysaccharides from fruit shell of Camellia oleifera,provide a theoretical basis for the actual development and utilization of fruit shell of C.oleifera.According to the yield and content of polysaccharide,the purifcation effects of different solvents such as methanol,ethanol and acetone and ethanol aqueous solutions with different concentration gradients such as 50%,60%,70%,80%and 90%were compared.Then,5%,10%,15%,20%and 25%activated carbons were used to decolorize and purify polysaccharide,and the best purifcation process was selected.The in vitro antioxidant activities of polysaccharides purifed from fruit shell of C.oleifera were determine by DPPH radical scavenging ability,ABTS+radical scavenging ability and Fe^(3+)reducing ability.The results showed when purifed by 80%ethanol aqueous solution,the polysaccharide content in fruit shell of C.oleifera was 34.93%,the yield was 39.55%,and the IC50 values for ABTS+radical and DPPH radical were 45.08μg/mL and 68.80μg/mL,respectively.When the concentration was 2 mg/mL,the reduction ability of iron ion was the strongest;When purifed by 80%ethanol combined with 10%activated carbon decolorization,the polysaccharide content in fruit shell of C.oleifera was 37.10%and the yield was 16.31%;The IC50 values of ABTS+radical and DPPH radical were 57.80μg/mL and 38.61μg/mL,respectively.When the concentration was 2 mg/mL,the reduction ability of iron ion was the strongest.The results from this study indicated that polysaccharide of C.oleifera possessed a strong anti-radical activity and might be an effective oxidation resistance treatment in the medical and food industries.
