摘 要:优选贯叶连翘精油的超临界CO_(2)提取工艺条件,并对其抗氧化活性进行研究。以精油提取率为评价指标,将单因素试验和响应面Box-Behnken试验设计法相结合,对影响精油提取率的因素,即提取温度、提取压力、CO_(2)体积流量、提取时间等进行考察,并采用多元线性方程及二项式方程拟合,以确定优化组合参数;以抗坏血酸(VC)为对照,对贯叶连翘精油清除DPPH·及·HO能力进行测试。最佳提取工艺参数为提取温度40℃、提取压力25MPa、CO_(2)体积流量25 L/h、提取时间58 min。在此条件下重复提取3次,贯叶连翘精油的平均提取率为0.72%,与理论预测0.77%较为接近。对DPPH·和·HO的IC50分别为0.777 mg/m L和0.953 mg/m L。优化后的贯叶连翘精油提取工艺稳定、可行,所得精油具有良好的抗氧化作用。试验结果为贯叶连翘抗氧化产品的综合开发提供理论依据,也为其他植物精油的提取和抗氧化性研究提供参考。To optimize the supercritical CO_(2) extraction process of essential oil from Hypericum perforatum L.and to explore the antioxidant activity of the extracts,taking the extraction rate of Hypericum perforatum L.as the index,extraction temperature,extraction pressure,CO_(2) fiow rate and extraction time on the extraction yields of essential oil from Hypericum perforatum L.were investigated by single factor experiment combined with response surface method of Box-Behnken design.Using a predictive continuous variable surface model and multiple linear equations to ft the experiment data,determine the optimal supercritical CO_(2) extraction condition.The ability of essential oil to scavenge DPPH·and·HO was tested with anticirculating acid(VC)as the reference substance.The results showed that the optimum extraction parameters were reached as following:extraction temperature 40℃,extraction pressure 25 MPa,CO_(2) fiow rate 25 L/h and extraction time 58 min,respectively.Under the above extraction condition for three times,the average extraction yield of essential oil was 0.72%,which was close to the theoretical prediction of 0.77%.This optimized process was feasible and reliable,and essential oil extracted had a good antioxidant effect.The experiment provided a research reference for the further development of antioxidant products from Hypericum perforatum essential oil products.It also provided a theoretical basis for extraction and antioxidant study of volatile oil from other plants.
