摘 要:目的探究寄主和产地对肉苁蓉6种苯乙醇苷和粗多糖含量的影响,并评价抗氧化活性。方法采用HPLC法、苯酚-硫酸法测定肉苁蓉6种苯乙醇苷(松果菊苷、肉苁蓉苷A、管花苷A、毛蕊花糖苷、异毛蕊花糖苷、2’-乙酰基毛蕊花糖苷)和粗多糖含量,并测定其体外抗氧化活性(DPPH、ABTS清除能力和总抗氧化能力)。结果景泰、和田、阿克苏四翅滨藜-肉苁蓉6种苯乙醇苷总量分别较景泰梭梭-肉苁蓉提高了186.69%、214.64%、815.52%;随着样品浓度的增加,肉苁蓉醇提物浓度为2 mg/mL时,对DPPH、ABTS清除率均达到了90%以上;粗多糖浓度为10 mg/mL时,对DPPH、ABTS清除率分别达到了80%和99%以上;当样品浓度为2 mg/mL时,景泰梭梭-肉苁蓉、景泰、和田、阿克苏四翅滨藜肉苁蓉与VC总抗氧化能力分别为1.56、3.63、3.52、4.38、4.56μmol/mL。结论寄主和产地对肉苁蓉苯乙醇苷成分、粗多糖含量与抗氧化活性有显著性影响(P<0.05),且肉苁蓉抗氧化能力与样品浓度呈量效关系,四翅滨藜-肉苁蓉各方面表现均优于梭梭-肉苁蓉。这一研究结果为肉苁蓉作为天然的抗氧化剂和新寄主四翅滨藜-肉苁蓉的推广提供了参考价值。Objective To investigate the effect of different hosts and producing areas on the content of six phenylethanoid glyco-sides and crude polysaccharides in Cistanches deserticola and to evaluate the antioxidant activity.Methods HPLC and phenol-sulfuric acid method were used to determine the contents of six phenylethanoid glycosides(Echinacoside,Cistanoside A,Tubulo-side A,Verbascoside,Isoacteoside,2'-Acetylacteoside)and crude polysaccharides in C.deserticola,then the antioxidant activ-ities in vitro(DPPH、ABTS scavenging capacity and total antioxidant capacity)were detected.Results The total amount of six phe-nylethanoid glycosides in Atriplex canescens-C.deserticola from Jingtai,Hotan and Aksu increased by 186.69%,214.64%and 815.52%,respectively,compared with Haloxylon ammodendron-C.deserticola from Jingtai;With increasing sample con-centration,the alcoholic extract of C.deserticola achieved more than 90%clearance of DPPH and ABTS at a concentration of 2mg/mL;The crude polysaccharides at a concentration of 10 mg/ml achieved over 80%and 99%clearance of DPPH and ABTS,respectively;When the sample concentration is 2mg/mL,the total antioxidant capacities of H.amodendron-C.deserticola in Jingtai,A.canescens-C.deserticola in Jingtai,Hotan and Aksu with VC were 1.56,3.63,3.52,4.38 and 4.56μmol/mL,respectively.Conclusion Host and origin had significant effects(P<0.05)on phenylethanoid glycosides composition,crude polysaccharides content and antioxidant activity of C.deserticola,the antioxidant capacity of C.deserticola was correlated with the concentration of the sample,A.canescens-C.deserticola was superior to H.ammodendron-C.deserticola in all respects.The results of this study provide a reference value for the promotion of C.deserticola as a natural antioxidant and A.canescens as a new host for C.deserticola.
