摘  要:以紫苏叶为原料,采用纤维素酶和果胶酶优化紫苏叶多糖提取的工艺,并研究其抗氧化活性。紫苏叶经过粉碎,采用单因素试验,选择纤维素酶添加量、果胶酶添加量、酶解时间和酶解温度四个因素,利用正交试验法优化紫苏叶多糖的提取工艺条件。结果表明,正交试验确定的紫苏叶多糖提取的最佳工艺参数为纤维素酶添加量2.5%,果胶酶添加量2.5%,酶解时间60 min,酶解温度50℃,紫苏叶多糖提取率最大,可达到3.01%。紫苏叶多糖对DPPH自由基和羟基自由基均具有较好的清除能力。以期为紫苏叶产品深加工提供理论依据。Taking Perilla leaf as raw material,cellulase and pectinase were used to optimize the extraction process of polysaccharides from Perilla leaf,and their antioxidant activities were studied.After crushing Perilla leaves,the content of cellulase,the content of pectinase,the time of enzymatic hydrolysis and the temperature of enzymatic hydrolysis were selected as the main factors on the basis of single factor test,and the orthogonal experimental was used to determine the maximum extraction rate of polysaccharide from Perilla leaves.The results showed that the optimal extraction parameters were cellulase content 2.5%,pectinase content 2.5%,enzymolysis time 60 min,enzymolysis temperature 50℃,and the maximum extraction yield of polysaccharides from Perilla leaves could reach 3.01%.Perilla polysaccharide has good scavenging ability on DPPH free radical and hydroxyl free radical.To provide theoretical basis for deep processing of Perilla leaf products.
