恒温荧光PCR法与国标法在单增李斯特氏菌检验能力验证中的应用与分析

摘  要:通过参加单增李斯特氏菌检验能力验证,促进实验室单增李斯特氏菌的检测能力持续提高,确保检测结果准确、可靠。同时,探索恒温荧光PCR法在单增李斯特氏菌检测工作中的可行性。对能力验证编号为20-E323和20-P712的两组肉类冻干粉样品,使用恒温荧光PCR与国家标准《食品安全国家标准食品微生物学检验单核细胞增生李斯特氏菌检验》(GB 4789.30—2016)进行单核细胞增生李斯特氏菌的检测。结果显示,使用恒温荧光PCR法检出20-E323、20-P712为阳性结果,与GB 4789.30—2016国家标准方法检出的阳性结果一致,证明20-E323、20-P712为单核细胞增生李斯特氏菌,能力验证结果为满意。本实验室具备单增李斯特氏菌的检测能力,且结果可靠。本次能力验证中,恒温荧光PCR法具有与国标法同等的检出率,为实验室快速检测单增李斯特氏菌提供依据。To improve the detection ability of Listeria monocytogenes in the laboratory and ensure the accuracy and reliability of the test results by participating in the proficiency testing of Listeria monocytogenes.At the same time,the feasibility of isothermal fluorescence PCR in the detection of Listeria monocytogenes was explored.According to the isothermal fluorescence PCR and"National food safety standards-Food microbiological analysis-Detection of Listeria monocytogenes"(GB 4789.30-2016),the two groups of samples with number 20-E323 and 20-P712 were qualitatively detected.The positive results of 20-E323 and 20-P712 were detected by isothermal fluorescence PCR,which was consistent with the results of GB 4789.30-2016 national standard method.It was proved that 20-E323 and 20-P712 were Listeria monocytogenes.The test results were all.The laboratory has the ability to detect Listeria monocytogenes,and the results are reliable.In the proficiency testing of Listeria monocytogenes,the detection rate of isothermal fluorescence PCR was the same as that of the national standard method,which provided the basis for the rapid detection of Listeria monocytogenes.
