摘 要:目的:以复方石斛(铁皮石斛-党参-黄芪-黄精=2∶3∶3∶2)为原料进行总黄酮的提取。方法:通过单因素实验和响应面法对提取工艺进行优化,并测定提取物体外抗氧化活性和体内降血糖活性。结果:复方石斛总黄酮的最优提取工艺条件为乙醇浓度60%,超声时间50min,液料比为35,总黄酮提取率为2.59%,测定其对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-trinitrophenylhydrazine,DPPH)、2,2-联氮-双-3-乙基苯并噻唑啉-6-磺酸(2,2-Binazo-bis-3-ethyl Benzothiazole line-6-sulfonic acid,ABTS+)、羟基自由基的半数抑制率分别为7.189μg/ml、1.288μg/ml、1.468μg/ml。采用线虫实验测定其体内降糖活性,当提取物制备成120 mg/ml的溶液时,有较好的降糖活性。结论:研究制备的复方石斛提取液具有较好的抗氧化和降糖作用。Purpose:The total flavonoids were extracted from compound Dendrobium officinale(Dendrobium officinale-Codonopsis pilosula-Huangqi-Huangjing=2:3:3:2).Method:The extraction process was optimized through single factor experiments and response surface methodology,and the antioxidant activity in vitro and hypoglycemic activity in vivo of the extracts were measured.Results:The results showed that the optimal extraction process conditions for total flavonoids in compound Dendrobium officinale were ethanol concentration of 60%,ultrasound time of 50 minutes,liquid-solid ratio of 35,and total flavonoid extraction rate of 2.59%.The half inhibition rates of 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),2,2-biazo-bis-3-ethylbenzothiazoline-6-sulfonic acid(ABTS+),and hydroxyl radicals were determined to be 7.189μg/ml,1.288μg/ml,and 1.468μg/ml,respectively.The nematode experiment was used to determine its hypoglycemic activity in vivo,and when the extract was prepared into a 120 mg/ml solution,it had good hypoglycemic activity.Conclusion:The compound dendrobium extract prepared in the study has good antioxidant and hypoglycemic effects.
