摘 要:研究紫芝和和乳酸克鲁维酵母共发酵的最佳发酵工艺,以及其发酵物体外抗氧化和抗衰老活性。以1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率为响应值,在考察单因素试验的基础上,利用Box-Behnken响应面分析法确定最佳发酵工艺。通过DPPH和羟自由基清除能力,以及人成纤维细胞I型胶原蛋白(COL-I)和基质金属蛋白酶1(MMP-1)的生成量,分别评价紫芝发酵物抗氧化和抗衰老活性。紫芝发酵物的最佳发酵工艺为菌液添加量3.5%(质量分数),马铃薯葡萄糖水(PDB)培养基用量3.25%(质量分数),发酵时间68.5 h。得到的紫芝发酵物对DPPH和羟自由基清除能力的IC50,以质量分数计分别为0.309%和0.475%,可以显著促进COL-I的合成和减少MMP-1的生成,具有很强的抗氧化和抗衰老能力,优于传统的紫芝提取物。The optimal conditions of the co-fermentation of Ganoderma sinense and Kluyveromyces lactis were investigated,and the in vitro antioxidant and anti-aging activities of the Ganoderma.sinense ferment substances were further studied.Taking the scavenging capacity of 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)as the response value,the best fermentation process was determined by Box-Behnken response surface analysis on the basis of single factor experiment.The antioxidant and anti-aging activities of Ganoderma sinense ferment substances were evaluated by the scavenging capacity of DPPH and hydroxyl radicals,and the production of type I collagen(COL-I)and matrix metalloproteinase-1(MMP-1)in human skin fibroblasts,respectively.The optimal fermentation process of Ganoderma sinense ferment substances was as follows:3.5%(mass fraction)of bacterial liquid,3.25%(mass fraction)of potato dextrose water(PDB)medium and 68.5 h of fermentation time.The IC50 of Ganoderma sinense ferment substances for DPPH and hydroxyl radicals scavenging capacity were 0.309%and 0.475%by mass fraction,respectively.Ganoderma sinense ferment substances could significantly promote the synthesis of COL-I and reduce the production of MMP-1.It has much stronger antioxidant and anti-aging activities,compared to those of Ganoderma sinense extract.
