摘  要:以哈尔滨市工厂化生产的黑木耳菌包中分离纯化的1株病原菌“ZJ-01”为试材,采用形态学和分子生物学方法鉴定“ZJ-01”,研究了碳源、氮源、pH和温度对“ZJ-01”菌丝生长速度和长势的影响,以期为病原菌“ZJ-01”的防治提供参考依据。结果表明:“ZJ-01”菌落和菌丝体形态特征与白囊耙齿菌(Irpexlacteus)相似。ITS序列与白囊耙齿菌同源性最高,为99.30%~99.47%,系统发育树分析发现“ZJ-01”与I.lacteus聚在同一个分支上,其中与I.lacteus strain xsd08033(EU918701.1)亲缘关系最近。结合形态学和ITS序列结果将“ZJ-01”归属为白囊耙齿菌。柯赫氏法则验证表明“ZJ-01”为引起黑木耳菌包污染的病原菌。在试验范围内白囊耙齿菌“ZJ-01”菌丝体生长的最佳碳源为麦芽糖,氮源为酵母浸粉,pH为5~6,温度为30℃。The pathogenic‘ZJ-01',isolated and purified from Auricularia heimuer bag production factory in Harbin,was selected as the experimental material.The morphological and molecular biology methods were used to identify taxonomic status of‘ZJ-01'.The effects of carbon source,nitrogen source,pH,and temperature on the mycelium growth rate and growth status of‘ZJ-01'were studied,in order to provide reference for the prevention and control of‘ZJ-01'.The results showed that the colony and mycelium morphology of‘ZJ-O1'was similar to those of Irper lacteus.The ITS sequence homology of‘ZJ-01'with I.lacteuswas from 99.30%to 99.47%.'ZJ-01'had the closest genetic relationship with I.lacteus strain xsd08033(EU918701.1).ZJ-01'was identified belong to I.lacteus based on morphology and ITS sequence analysis.Koch's postulates proved that‘ZJ-01'was the pathogen that caused contamination of A.heimuer bag.Researches on growth characteristics showed that the best carbon source for'ZJ-01'mycelial growth was maltose,and the nitrogen source was yeast extract powder,pH of 5 to 6,and temperature of 30℃.
