摘 要:实验采用邻苯二甲醛和丹磺酰氯两种柱前衍生高效液相色谱法测定奶粉中牛磺酸的含量,并比较了两种方法的优劣。样品用水溶解除去蛋白质后,上清液用邻苯二甲醛和丹磺酰氯衍生,衍生产物经C18反相色谱柱分离,采用高效液相色谱法测定,以外标法定量。两种衍生化方法灵敏度好、分离度高,均可用于奶粉中牛磺酸的测定,但邻苯二甲醛法具有样品前处理快、分析时间短、干扰少等优点,是较理想的牛磺酸测定方法。The content of taurine in milk powder were determined by high performance liquid chromatography with two kinds of pre-column derivatization by o-phthalaldehyde dansyl chloride.The advantages and disadvantages of the two methods were compared.After the samples were dissolved with water to remove the protein,the supernatant was derivatized with o-phthalaldehyde and dansyl chloride.The derivatives were separated by C18 reversed-phase column and determined by HPLC and quantified by external standard method.The two derivatization methods have good sensitivity and high resolution and can be used for the determination of taurine in milk powder.However,the o-phthalaldehyde method has the advantages of fast sample preparation,short analysis time and less interference,and is an ideal taurine determination method.
