摘  要:通过单因素试验、正交试验对乳双歧杆菌BL-99的培养基组成和培养条件进行了优化研究。结果表明,最优培养基组成为:蛋白胨10g/L、酵母浸粉15g/L、葡萄糖40g/L;最佳培养条件为:pH6.0、湿度37℃、搅拌转速70rpm、接种量2%。In this study,Bifidobacterium lactis BL-99 was used as the research object.Single factor test and orthogonal test were applied to optimize the medium composition and culture conditions of high cell density fermentation on the basis of the modified MRS medium.The results showed that the optimal medium composition of B.lactis BL-99 was:peptone 10g/L,yeast extract 15g/L,glucose 40g/L,anhydrous sodium acetate 5g/L,potassium citrate 2.5g/L,diammonium phosphate 1.5g/L,magnesium sulfate heptahydrate 1g/L,sodium erythorbate 1g/L,manganese sulfate monohydrate 0.19g/L,Tween-801g/L;The optimal culture conditions were as follows:pH 6.0,fermentation temperature 37℃ , stirring speed 70rpm, inoculum 2%. Under these optimized conditions, the numberof viable bacteria of B. lactis BL-99 reached 2.1×1010CFU/mL, which were 13.1 times higher than that of beforeoptimization. The results will significantly improve the industrial production efficiency of B. lactis BL-99.
