摘  要:本文研究了4种来源的胆碱酯酶酶制剂稳定性和保存条件,底物、显色剂等酶抑制法检测条件,对比了酶抑制法与气相色谱法、农药速测卡对蔬菜中有机磷的检测效果。实验表明胆碱酯酶酶制剂于-18℃下保存20 d,其活力较为稳定,之后活力下降;底物和显色剂于4℃下保存,稳定性好。本实验酶抑制法所测定的有机磷残留比标准气相色谱法的准确度稍低,样品中农药残留浓度测定值的绝对误差值在0~0.029 mg/kg,相对误差值在0~17.6%;与市售的农药速测卡相比,其对农药残留浓度的检出限比农药速测卡的检出限低,其中对甲胺磷的检出限为0.10 mg/kg,毒死蜱的检出限为0.20 mg/kg。本实验方法为实际样品的大批量的检测提供了参考。The stability and preservation conditions of cholinesterase enzyme preparations from four sources were studied in this paper,and the detection conditions of substrate,chromogenic agent and other enzyme inhibition methods were optimized.The detection effects of enzyme inhibition methods were compared with gas chromatography and pesticide rapid test cards on organophosphorus in vegetables.The results showed that the activity of cholinesterase was relatively stable when stored at-18℃ for 20 days,and then decreased.The substrate and chromogenic agent are stored at 4℃ with good stability.The enzyme inhibition method used in this experiment has a higher accuracy in determining organophosphorus residues,which is slightly lower than the accuracy of gas chromatography.The absolute error value of pesticide residue concentration in the sample is between 0 to 0.029,and the relative error value is between 0 to 17.6%.Compared with commercially available rapid test cards,the detection limit of pesticide residues was lower than that of rapid test cards,with a detection limit of 0.10mg/kg for methamidophos and 0.20mg/kg for chlorpyrifos.This experimental method provides a basis for the detection of large quantities of actual samples.
