摘 要:本试验以荷叶为原料,利用超声波辅助碱性蛋白酶酶解荷叶制备小分子多肽。选用底物浓度、酶解温度、酶解pH、酶解时间四个因素进行单因素试验。再通过响应面试验对提取荷叶多肽工艺进行优化。最后以多肽浓度为标准,得到最佳酶解得荷叶多肽的条件为:底物浓度4.07%、酶解温度54.96℃、酶解pH 8.88、酶解时间5.19 h。在上述条件下,得到多肽浓度为8.98×10^(-3) mg/mL。Using lotus leaf as raw material,small molecular polypeptides were prepared by enzymatic hydrolysis of lotus leaf assisted by alkaline protease.The four factors of substrate concentration,enzymolysis temperature,enzymolysis pH and enzymolysis time were tested by single factor test.The extraction process of polypeptide from lotus leaf was optimized by response surface test afterwards.Finally,the optimum conditions for enzymatic hydrolysis of lotus leaf polypeptides were as follows:substrate concentration 4.07%,enzymatic hydrolysis temperature 54.96℃,enzymatic hydrolysis pH 8.88,and enzymatic hydrolysis time 5.19 h.Under the above conditions,the concentration of polypeptide was 8.98×10^(-3) mg/mL.
