摘 要:目的探讨长链非编码RNA(lncRNA)小核仁RNA宿主基因17(SNHG17)在肝细胞癌(HCC)发生发展中的作用。方法利用基因表达谱交互分析(GEPIA)数据库,分析HCC中SNHG17的表达及其与生存的关系;利用基因表达综合(GEO)数据库的数据进行验证,使用GEO2R、lnCAR和Metascape软件分析SNHG17相关性基因、构建可视化网络并进行GO和KEGG富集分析。根据通路利用多个GEO数据集筛选出细胞周期蛋白依赖性激酶4(CDK4);利用数据库平台StarBase、miRBD以及HMDD筛选出同时以SNHG17和CDK4为靶基因的microRNA(miRNA)。结果lncRNA SNHG17在HCC中的表达显著上升(P<0.05),其表达高的病人预后显著变差(P<0.05)。GO和KEGG富集分析结果显示,SNHG17与核蛋白复合物以及细胞周期相关。筛选出高度相关基因CDK4并筛选出以SNHG17和CDK4为靶标的miRNAs。结论SNHG17在HCC中表达增高,并可通过结合多种miRNA发挥竞争性内源RNA的功能,对CDK4发挥调控作用。Objective To investigate the role of the long non-coding RNA(lncRNA)small nucleolar RNA host gene 17(SNHG17)in the development and progression of hepatocellular carcinoma(HCC).Methods GEPIA database was used to analyze the expression of SNHG17 in HCC and its association with survival;the data in GEO database were used for validation;GEO2R,lnCAR,and Metascape were used to analyze SNHG17-related genes,construct a visualized network,and perform GO and KEGG enrichment analyses.Based on related pathways,multiple GEO datasets were used to screen for CDK4,and StarBase,miRBD,and HMDD database platforms were used to obtain the microRNAs(miRNAs)targeting both SNHG17 and CDK4.Results There was a significant increase in the expression of lncRNA SNHG17 in HCC(P<0.05),and the patients with high SNHG17 expression tended to have a poorer prognosis(P<0.05).GO and KEGG enrichment analyses showed that SNHG17 was associated with nuclear protein complex and cell cycle.The highly related gene CDK4 was obtained,as well as the miRNAs targeting SNHG17 and CDK4.Conclusion There is an increase in the expression of SNHG17 in HCC,and SNHG17 can play the role of competitive endogenous RNA by binding to a variety of miRNAs,thereby exerting a regulatory effect on CDK4.
