摘　　要：目的本研究旨在探讨环状RNA(circRNA)EPB41L2通过调控miR-1270/TFRC轴对口腔鳞癌(Oral squamous cell carcinoma,OSCC)细胞的增殖、迁移和侵袭的影响。方法qRT-PCR分析circRNA EPB41L2、miR-1270和TFRC的表达水平,克隆形成、Transwell实验分别分析OSCC细胞增殖、迁移和侵袭,蛋白质免疫印迹法分析上皮-间充质转化(epithel-mesenchymal transition,EMT)标记物的表达。双荧光光素酶和RIP实验以探索circRNA EPB41L2、miR-1270和TFRC之间的相互作用。结果circRNA EPB41L2在OSCC细胞中上调,细胞功能实验表明,敲低circRNA EPB41L2抑制了细胞增殖、迁移和侵袭。此外,机制研究表明circRNA EPB41L2可以与miR-1270结合并上调其靶基因TFRC,从而促进OSCC细胞的EMT。结论本研究的结果证明了circRNA EPB41L2在OSCC肿瘤发生和转移中的作用,circRNA EPB41L2通过OSCC中miR-1270/TFRC轴发挥其促癌作用。Objective The purpose of this study was to investigate the effect of circular RNA(circRNA)EPB41L2 on the proliferation,migration and invasion of oral squamous cell carcinoma(OSCC)cells by regulating the miR-1270/TFRC axis.circRNA EPB41L2 has been reported to be abnormally expressed in cancer,but its role in tumor growth and metastasis remains unclear.Method The expression levels of circRNA EPB41L2,miR-1270 and TFRC were analyzed by qRT-PCR.The proliferation,migration and invasion of OSCC cells were analyzed by clone formation and Transwell assay,respectively.The expression of epithelial-mesenchymal transition(EMT)markers was analyzed by Western blotting.Dual luciferase and RIP assays were performed to explore the interaction between circRNA EPB41L2,miR-1270 and TFRC.Result circRNA EPB41L2 was up-regulated in OSCC cells,and cell function experiments showed that circRNA EPB41L2 knockdown inhibited cell proliferation,migration,and invasion.Moreover,mechanism studies have shown that circRNA EPB41L2 can bind to miR-1270 and up-regulate its target gene TFRC,thereby promoting EMT of OSCC cells.Conclusion The results of this study demonstrated the role of circRNA EPB41L2 in OSCC tumorigenesis and metastasis,and circRNA EPB41L2 plays its role in promoting cancer through the miR-1270/TFRC axis in OSCC.
