摘 要:目的:针对目前国标法未涉及微生物法测定饮料中维生素B_(12)的情况,通过验证预包被微孔板式微生物法测定饮料中维生素B_(12)的可行性和可靠性,为实验室开展该项目和创立新方法提供有力支持。方法:利用德氏乳杆菌生长依赖维生素B_(12)的特性,根据德氏乳杆菌生长状况与维生素B_(12)含量呈正相关关系,通过测定样品吸光度值与标准曲线进行比对得出维生素B_(12)的含量。结果:此方法操作简便、线性良好,R^(2)可达0.998 3,回收率在95.5%~104.6%,重复实验10次,品牌1、2的RSD值分别为1.367%和0.597%,试剂盒质量稳定性较好。结论:此方法适用于饮料中维生素B_(12)的测定,为实验室开展该项目和新方法的研究奠定了基础。Objective:In view of the fact that the microbiological method for the determination of vitamin B_(12) in beverages is not covered by the current national standard method,the feasibility and reliability of the pre-coated microplate microbiological method for the determination of vitamin B_(12) in beverages were verified,which provided a strong support for the laboratory to carry out this project and create a new method.Method:Utilizing the property that the growth of Lactobacillus delbrueckii is dependent on vitamin B_(12),and according to the positive correlation between the growth condition of Lactobacillus delbrueckii and the content of vitamin B_(12),the content of vitamin B_(12) was obtained by measuring the absorbance value of the sample and comparing with the standard curve.Result:The method is easy to operate and has good linearity with R^(2) up to 0.9983,the recoveries were in the range of 95.5%~104.6%,and the experiments were repeated for 10 times,the RSD values of brands 1 and 2 were 1.367%and 0.597%,respectively,and the quality stability of the kit was good.Conclusion:This method is suitable for the determination of vitamin B_(12) in beverages,and lays the foundation for the laboratory to carry out the research of this program and new methods.
