玉米肽保护急性酒精性肝损伤的作用研究

摘　　要：目的研究玉米低聚肽及玉米肽糙米胚芽片对于急性酒精性肝损伤是否具有保护作用。方法采用一次性灌胃14 mL·kg^(-1)的50%乙醇构建急性酒精性肝损伤小鼠模型;测定小鼠血清和肝脏组织中肝损伤指标AST、ALT、酒精代谢指标ADH、ALDH、脂质代谢/过氧化指标TG、MDA和GSH水平来评估肝损伤程度,采用HE染色评价肝脏病理情况。结果小鼠一次性灌胃大量酒精后,肝脏AST和ALT酶活显著升高、ADH和ALDH酶活降低,GSH水平下降,MDA和TG大量积累;病理切片观察到肝索紊乱,肝细胞脂肪性病变。玉米低聚肽和玉米肽糙米胚芽片各剂量组均能显著改善以上指标和肝脏病理损伤,呈现一定量效关系。结论一次性大量饮酒会造成小鼠肝功能异常,玉米低聚肽作为一种功能性小分子混合物,具有良好的改善急性酒精性肝损伤的能力,玉米肽糙米胚芽片在玉米低聚肽的基础上增加了牛磺酸等功能性成分,同样具有良好的缓解小鼠急性酒精肝损伤的能力。OBJECTIVE To investigate whether corn oligopeptides and Corn peptide and brown rice germ tablets have protective effects on acute alcoholic liver injury.METHODS The mice model of acute alcoholic liver injury was established by intragastric administration of 14 mL·kg^(-1)of 50% ethanol at one time.The levels of liver damage indicators(AST and ALT),alcohol metabolism indicators(ADH and ALDH),and lipid metabolism/peroxidation indicators(TG,MDA and GSH)in the serum and liver tissue of mice were measured to evaluate the degree of liver damage.H&E stain was used to assess liver pathology.RESULTS The results showed that taking large amounts of alcohol increases the levels of AST and ALT,and decreases the levels of ADH,ALDH and glutathione in mice.MDA and TG accumulate in large amounts in the liver.Liver pathology sections reveal disorganized liver cords,massive balloon-like lesions,and widely distributed lipid droplets.Corn oligopeptides and Corn peptide and brown rice germ tablets can significantly liver pathological damage in a certain dose-effect relationship.CONCLUSION Corn oligopeptides,as a functional small molecule mixture,have good ability to improve acute alcoholic liver injury induced by one-time heavy drinking.Corn peptide and brown rice germ tablets,based on corn oligopeptides and adding functional ingredients such as taurine,also have a good ability to alleviate acute alcoholic liver injury in mice.
