摘 要:目的:探究苦碟子注射液中的多糖类成分理化性质、表征结构并分析其体外抗氧化活性能力。方法:采用醇沉法从苦碟子注射液中富集多糖,D101大孔吸附树脂法脱色;采用DEAE-52纤维素离子交换层析柱、Sephadex G-100纯化得到苦碟子多糖KDTD1_(1);采用PMP(1-苯基-3-甲基-5-吡唑啉酮)柱前衍生化、凝胶渗透色谱、红外光谱、甲基化法对KDTD1_(1)进行结构表征,并考察其清除羟基自由基、DPPH自由基及ABTS自由基能力以评价KDTD1_(1)体外抗氧化活性。结果:KDTD1_(1)中性糖含量为90.5%,酸性糖含量为0.05%,蛋白质含量为0.02%,相对分子质量为1.2×10^(4),KDTD1_(1)由Rha、Glc、Gal和Xyl组成,摩尔质量比为1.5∶9.8∶1∶0.8;KDTD1_(1)主要包含8种糖残基,其中有3个末端,2个分支点,主要以→2)-Glcp-(1→及→4)-Glcp-(1→为主链。抗氧化试验结果显示当KDTD1_(1)质量浓度为1 mg·mL^(-1)时,对羟自由基、DPPH自由基、ABTS自由基的清除率分别为82.3%、82.4%、67.6%,并在0.1~1.0 mg·mL^(-1)范围内呈现一定量效关系。结论:从苦碟子注射液中分离得到具有支链的杂多糖KDTD1_(1)且具有一定的抗氧化活性,对进一步明确苦碟子注射液的药效物质基础打下基础。OBJECTIVE To investigate the physicochemical properties,characterize the structure and analyze the in vitro antioxidant activity of the polysaccharide components in Kudiezi injection.METHODS Polysaccharides were enriched from Kudiezi injection by alcohol precipitation and decolorized by D101 macroporous adsorption resin;the polysaccharide KDTD1_(1)was purified by DEAE-52 cellulose ion exchange chromatography column and Sephadex G-100;the structure of KDTD1_(1)was characterized by PMP(1-phenyl-3-methyl-5-pyrazolone)pre-column derivatization,gel permeation chromatography,infrared spectroscopy and methylation,and its ability to scavenge hydroxyl radicals,DPPH radicals and ABTS radicals was investigated to evaluate the in vitro antioxidant activity of KDTD1_1.RESULTS The polysaccharide KDTD1_(1)had a neutral sugar content of 90.5%,an acid sugar content of 0.05%,a protein content of 0.02%,and a relative molecular weight of 1.2×10^(4).KDTD1_(1)was composed of Rha,Glc,Gal,and Xyl,with a molar mass ratio of 1.5∶9.8∶1∶0.8;KDTD1_(1)mainly contained eight sugar residues,among which there were 3 termini and 2 branching points,mainly with→2)-Glcp-(1→and→4)-Glcp-(1→as the main chain.The results of the antioxidant test showed that when the mass concentration of KDTD1_(1)was 1 mg·mL~(-1),the scavenging rates of hydroxyl radical,DPPH radical,and ABTS radical were 82.3%,82.4%,and 67.6%,respectively,which showed a certain quantitative-effect relationship within the concentration of 0.1-1 mg·mL~(-1).CONCLUSION Heteropolysaccharide KDTD1_(1)with branched chain and certain antioxidant activity was isolated from Kudiezi injection,which lays foundation for further clarification of the pharmacodynamic material basis of Kudiezi injection.
