摘 要:以疏花蔷薇花中总多酚、鞣花酸、浸膏得率的综合评分作为评价指标,采用单因素试验结合响应面法优化疏花蔷薇花总多酚的提取工艺参数,并测定所得提取物对DPPH自由基和ABTS+自由基的清除能力及其还原能力。结果表明,疏花蔷薇花总多酚最佳提取工艺参数为采用55%乙醇作为提取溶剂,料液比1∶25(g/mL),回流提取2次、90 min/次,所得提取物的浸膏得率为(51.36±0.78)%,总多酚含量为(141.36±2.07)mg/g,鞣花酸含量为(13.46±0.14)mg/g,与模型预测值相当。疏花蔷薇花提取物对DPPH自由基、ABTS+自由基清除率的半抑制浓度(the half maximal inhibitory concentration,IC_(50))值分别为0.02、0.03 mg/mL,还原能力较强。The comprehensive score of total polyphenols,ellagic acid,and extract yield from Rosa laxa Retz.flowers was used as the evaluation index.The extraction process parameters of total polyphenols from Rosa laxa Retz.flowers were optimized by a single factor experiment combined with the response surface method.The scavenging ability and reducing ability of the extracts to DPPH and ABTS+free radicals were determined,and the antioxidant activities were investigated in vitro.The optimal extraction process parameters of total polyphe⁃nols from Rosa laxa Retz.flowers were as follows:55%ethanol was used as extraction solvent;the ratio of solid to liquid was 1∶25(g/mL).Reflux extraction was performed twice for 90 min/time.The extract yield was(51.36±0.78)%;the total polyphenol content was(141.36±2.07)mg/g,and the ellagic acid content was(13.46±0.14)mg/g,which were equivalent to the value predicted by the model.The half maximal inhibitory concentration(IC_(50))values of DPPH and ABTS+free radical scavenging rate of the extract from Rosa laxa Retz.flowers were 0.02、0.03 mg/mL,respectively,and its reducing ability is strong.
