摘 要:芽孢杆菌来源的纤维素酶具有耐热耐碱等优良催化特性,故而受到了广泛关注。本研究利用筛选的芽孢杆菌B55发酵生产羧甲基纤维素酶,在确定碳源、氮源、金属盐、羧甲基纤维素添加量、接种量等对菌株产酶影响的基础上,进行正交试验优化以确定最佳发酵培养工艺。研究结果表明:最佳培养工艺的配方为30 g/L甘油、5 g/L葡萄糖、3 g/L大豆蛋白胨、16 g/L酵母浸粉、4 g/L羧甲基纤维素、3 g/L磷酸氢二钾,3 g/L磷酸二氢钠,pH 7.0,在4%(V/V)接种量、培养温度32℃的条件下,发酵48 h获得最大酶活力401.34 U/mL,具备规模生产的应用潜力。Cellulase from Bacillus has excellent catalytic properties such as heat and alkali resistance,so it has been attracted wide attention.In this paper,carboxymethyl cellulase(CMCase)was produced with the screened Bacillus sp.B55.On the basis of determining the effect of carbon source,nitrogen source,metal salt,carboxymethyl cellulose addition and inoculation amount on the CMCase production of the strain B55,the fermentation cultural process was further optimized by orthogonal experiment design.The results indicated the optimal medium was 30 g/L glycerine,5 g/L glucose,3 g/L soybean peptone,16 g/L yeast powder,4 g/L CMC,3 g/L K2HPO_(4),3 g/L NaH_(2)PO_(4),pH 7.0,and then 401.34 U/mL of the maximum CM Case activity was obtained at 48 h under the conditions of 4%(V/V)inoculums and culture temperature of 32℃,so which had potential for scale production and application.
