摘 要:对黑曲霉发酵麸皮产木聚糖酶、阿魏酸酯酶活性与产总阿魏酸量的相关性进行研究,运用Box-Behnken实验及响应面分析,对麸皮培养基配方进行优化,提取黑曲霉发酵麸皮产总阿魏酸,并检测其抑菌效果。实验结果显示,木聚糖酶和阿魏酸酯酶活性与麸皮总阿魏酸释放量呈正相关。黑曲霉发酵麸皮产总阿魏酸工艺参数优化组合为:麸皮10 g,料液比为1∶1.4,麦芽糖、尿素、磷酸二氢钾在所加溶液中的质量浓度分别为6.42、2.00、0.11 g/L,黑曲霉种子液接种量为1.0 mL,30℃发酵96 h,麸皮总阿魏酸释放量为2.72 mg/g。以体积分数75%乙醇为溶剂,黑曲霉发酵麸皮产总阿魏酸提取量最大,为2.92 mg/g。用体积分数80%、75%乙醇提取的总阿魏酸分别对大肠杆菌、金黄色葡萄球菌的抑菌效果最佳。The correlation between xylanase,ferulic acid esterase activity and total ferulic acid production from bran fermented by Aspergillus niger was studied experimentally.The formula of bran culture medium was optimized by Box-Behnken experiment and response surface analysis,the total ferulic acid produced by Aspergillus niger fermented bran was extracted and its antibacterial effect was tested.The experimental results indicated that the activities of xylanase and ferulic acid esterase were positively correlated with the total ferulic acid release amounts from fermented bran.The optimal combination of technological parameters for producing total ferulic acid from wheat bran by Aspergillus niger fermentation was as follows:wheat bran was 10 g,the ratio of material to liquid was 1∶1.4,the concentrations of maltose,urea and potassium dihydrogen phosphate in the added solution were 6.42 g/L,2.00 g/L and 0.11 g/L respectively,the inoculation amount of Aspergillus niger seed solution was 1.0 mL,and the total ferulic acid released amounts from fermented wheat bran was 2.72 mg/g after fermentation at 30℃for 96 h.The 75%ethanol was used as the extraction solution,the total ferulic acid extracted from bran fermented by Aspergillus niger was the largest,being 2.92 mg/g.The total ferulic acid extracted with 80%and 75%ethanol had the best inhibitory effects on Escherichia coli and Staphylococcus aureus,respectively.
