摘 要:建立基于免疫层析技术的食品中米酵菌酸的快速检测方法。对银耳、河粉及玉米粉等代表性基质样品采用胶体金免疫层析法进行检测。优化了快速检测的前处理流程,并考察了胶体金免疫层析商品化产品在典型基质中的检出限、灵敏度、假阳性率、假阴性率、交叉反应率、温度耐变性、抗基质干扰能力、准确度和重复性等指标。结果表明,优化后的前处理条件使该方法检出限达到15μg/kg,灵敏度为100%,假阳性率和假阴性率均为0,脂肪酸类结构类似物和真菌毒素类物质交叉反应率为0。同时,该方法对检测环境温度不敏感且抗食品基质干扰能力强。另外,该方法具有较高准确度且重复性良好,与参比方法结果具有一致性。该方法易操作、用时少、准确性高,检出限低,适合批量样品的现场快速筛查。A rapid detection method for bongkrekic acid in food based on immunochromatography technology was established.The colloidal gold immunochromatography assay was used to detect the representative matrix samples of tremella,rice noodle and corn flour.The pretreatment steps of rapid detection were optimized,and the detection limit,sensitivity,false positiverate,false negative rate,cross reaction rate,temperature variability,anti-matrix interference ability,accuracy and repeatability of colloidal gold immunochromatography products in typical matrices were investigated.The limit of detection was reached 15μg/kg with a sensitivity rate of 100%using the optimized pretreatment conditions.In the meantime,both the false positive and negative rate were 0,the cross-reaction rate of fatty acid structural analogues and mycotoxins was 0 as well.At the same time,this method was not sensitive to the detection of ambient temperature and had strong resistance to food matrix interference.In addition,it had high accuracy and good repeatability,which was consistent with the results of the reference method.The method was simple,rapid,accurate and low detection limit,which was suitable for rapid on-site screening of batch samples.
