摘 要:共轭亚油酸(CLA)是亚油酸位置和构型异构体的总称,在保健品中已经有了共轭亚油酸的应用。为了进一步提高共轭亚油酸在食品应用中的安全性,区分共轭亚油酸中作用不同的物质。采用层析中的柱色谱法(又称柱层析),用尿素和硅胶制备一种全新的柱层析材料,定向吸附当前共轭亚油酸产品中的杂质成分。在径高比为1∶9,正己烷为洗脱剂的条件下,一次柱层析即可将共轭亚油酸含量提高6%以上,且两种全反式构型的共轭亚油酸异构体(包括t9,t11-CLA和10t,t12-CLA)含量下降明显;在两次柱层析的产品中,两种全反式结构的共轭亚油酸异构体含量下降至0.2%左右,有效增加了共轭亚油酸在应用中的安全性。本研究为纯化共轭亚油酸提供了新的方法,并且降低了共轭亚油酸应用在食品中潜在的风险。Conjugated linoleic acid(CLA)is a generic term for the positional and conformational isomers of linoleic acid,which have been used in nutraceuticals.To further improve the safety of conjugated linoleic acid in food applications,substances with different roles in the conjugated linoleic acid were distinguished.Using column chromatography(also known as column chromatography),a novel column chromatography material was prepared using urea and silica gel for selectively adsorption of impurity components in conjugated linoleic acid products.Under the conditions of 1∶9 diameter-to-height ratio and hexane as eluent,the content of conjugated linoleic acid could be increased by more than 6%in one column chromatography,and the content of two all-trans isomers of conjugated linoleic acid(including t 9,t11-CLA and t10,t12-CLA)decreased significantly;In the products subjected to the second column chromatography,the content of two all-trans structures of conjugated linoleic acid isomers decreased to about 0.2%,which effectively increased the safety of conjugated linoleic acid in applications.This study provides a new method for the purification of conjugated linoleic acid and reduces the potential risk of conjugated linoleic acid application in food.
