摘 要:采用梯度稀释涂布法从常年落叶的土壤中筛选出3株具有降解β-胡萝卜素能力的菌株,通过分光光度法测定发酵液中β-胡萝卜素的降解率,进而对发酵液中的产物进行GC-MS分析,筛选出目标菌株TR-3,其降解β-胡萝卜素的能力在3株中最强,降解率达86.65%。革兰氏染色和光学显微镜观察结果表明,该菌株为革兰氏阴性杆菌;16S rDNA测序及聚类发育分析结果表明,菌株TR-3为肠杆菌属;气质联用(GC-MS)法分析发现,其对β-胡萝卜素降解产物中的成分主要为仲辛酮(37.69%)、苯乙醇(13.88%)、磷酸三丁脂(5.55%)、癸酸乙酯(5.54%)、β-紫罗兰酮(4.80%)及月桂酸乙酯(3.73%)等致香物质。In this experiment,three strains with the ability to degradeβ-carotene were preliminarily screened from the perennial deciduous soil by gradient dilution coating method.The degradation rate ofβ-carotene in the fermentation broth was determined by spectrophotometer.The products in the fermentation broth were analyzed by GC-MS,and the target strain TR-3 was screened out.The ability to degradeβ-carotene was the strongest among the three strains,and the degradation rate was 86.65%.The results of Gram staining and optical microscopy showed that the strain was Gram-negative bacilli.The results of 16S rDNA sequencing and cluster development analysis showed that strain TR-3 was Enterobacter.The degradation products were preliminarily analyzed by gas chromatography-mass spectrometry(GC-MS).The products were mainly aromatic substances such as 2-octanone(37.69%),phenylethyl alcohol(13.88%),tributyl phosphate(5.55%),ethyl decanoate(5.54%),β-ionone(4.80%)and ethyl laurate(3.73%).
