摘 要:【目的】采用响应面法优化超声波辅助提取地涌金莲多酚的工艺条件,评价其抗氧化活性,以期为地涌金莲的开发利用提供参考依据。【方法】以地涌金莲花瓣为原料,在单因素试验的基础上,依据Box-Behnken中心组合原理选择超声波功率(X_(1))、提取温度(X_(2))、提取时间(X_(3))和料液比(X4)为试验因素,多酚提取量(Y)为响应值进行响应面试验,优化得到超声波辅助提取的最佳工艺条件;并以清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)阳离子自由基的能力及总抗氧化能力(FRAP)为指标,评价地涌金莲多酚提取物的抗氧化活性。【结果】各因素对地涌金莲多酚提取量的影响排序:X_(2)>X_(3)>X_(1)>X4;X_(1)X_(2)、X_(1)X_(3)的交互作用对多酚提取量的影响极显著(P<0.01),X_(2)X_(3)的交互作用对多酚提取量的影响显著(P<0.05)。模型方程优化为Y=100.73-4.87X_(1)X_(2)-5.03X_(1)X_(3)-4.09X_(2)X_(3)-11.13X_(1)^(2)-9.06X^(2)_(2)-12.05X_(3)2-10.46X42,此时的地涌金莲多酚提取最佳工艺参数为超声波功率360 W、提取温度55°C、提取时间30 min、料液比1∶20(g/mL),在此条件下的地涌金莲多酚提取量为10.51 mg/g,与理论值(10.08 mg/g)接近。抗氧化试验结果显示,地涌金莲多酚提取物清除DPPH自由基和ABTS阳离子自由基的半抑制浓度(IC_(50))分别为20.03和39.68μg/mL,FRAP值为47.540μg/mL,且抗氧化能力高于槲皮素。【结论】超声波辅助提取的地涌金莲多酚具有良好的抗氧化活性,可开发为一种安全、天然、可食用的抗氧化剂。【Objective】The response surface method was used to optimize the process conditions of ultrasonic-assisted extraction of polyphenols from Musella lasiocarpa(Franch.),and the antioxidant activities of M.lasiocarpa poly‐phenol extract(MLPE)were further evaluated,to provide theoretical support for further development and utilization of M.lasiocarpa.【Method】Ultrasonic power(X_(1)),extraction temperature(X_(2)),extraction time(X_(3))and solid-liquid ratio(X4)were selected as experimental factors,and polyphenol yield(Y)was the response value,response surface methodo-logy was conducted to optimize conditions of ultrasonic-assisted extraction from M.lasiocarpa petal on the basis of single factor experiments.The antioxidant activity of the polyphenol extract was evaluated by the scavenging ability of 1,1-diphe‐nyl-2-trinitrophenylhydrazine(DPPH)free radicals and 2,2-diazo-bis(3-ethyl-benzothiazole-6-sulfonic acid)diamiam‐monium salt(ABTS)and total antioxidant capacity(FRAP).【Result】The effect of each factor on the extraction rate of polyphenol from the M.lasiocarpa was X_(2)>X_(3)>X_(1)>X4.The effect of interaction between X_(1)X_(2) and X_(1)X_(3) on the poly‐phenol extraction rate were extremely significant(P<0.01),while the effect of interaction effect of X_(2)X_(3) was significant(P<0.05).The optimized model equation was Y=100.73-4.87X_(1)X_(2)-5.03X_(1)X_(3)-4.09X_(2)X_(3)-11.13X_(1)2-9.06X_(2)^(2)-12.05X_(3)^(2)-10.46X42,and the optimal extraction process parameter for polyphenols from the M.lasiocarpa were ultrasonic power of 360 W,extraction temperature of 55℃,extraction time of 30 min,and liquid-solid ratio of 1∶20(g/mL).Under these op‐timal conditions,the extraction amount of polyphenols from M.lasiocarpa was 10.51 mg/g,which was consistent with the theoretical value of 10.08 mg/g.Results from antioxidant assays showed that the half maximal inhibitory concentration(IC_(50))values of DPPH and ABTS radical scavenging activity were 20.03 and 39.68μg/mL,respectively,with a FRAP values of 47
