摘 要:目的:研究下调泛素特异性肽酶18(USP18)对HepG2肝癌细胞生长和迁移的影响,并探讨其影响HepG2细胞恶性行为作用机制。方法:通过基因转染的方法构建低表达USP18的HepG2细胞株(Si-USP18)及其对照细胞株(Si-NC),分别利用细胞计数试剂-8(CCK-8)实验及细胞迁移(Transwell)实验来研究敲低USP18对HepG2细胞增殖和迁移的影响,并应用蛋白印记(WB)实验来研究敲低USP18对HepG2细胞mTOR通路相关蛋白表达的影响。结果:CCK-8实验表明,细胞培养24、48、72h后,与Si-NC组细胞比较,Si-USP18组细胞的增殖能力显著增强(P<0.05);Transwell实验表明,细胞培养24h和48h后,与Si-NC组细胞比较,Si-USP18组细胞的迁移能力显著增强(P<0.05);WB实验表明,与Si-NC组细胞比较,Si-USP18组细胞的磷酸化-哺乳动物雷帕霉素靶蛋白(p-mTOR)、B细胞淋巴瘤-2(Bcl-2)和细胞周期蛋白D1(cyclin D1)蛋白表达水平显著升高(P<0.05)。结论:下调USP18可通过mTOR信号通路促进肝癌细胞的恶性行为。Objective:To investigate the effect of downregulation of ubiquitin-specific protease 18(USP18)on the growth and migration of HepG2 hepatocarcinoma cells,and to explore its mechanism of effect on the malignant behavior of HepG2 cells.Methods:HepG2 cells with low USP18 expression(si-USP18)and its control cells(Si-NC)were constructed by gene transfection,the effects of USP18-deficiency on the proliferation and migration of HepG2 cells were investigated by cell counting reagent-8(CCK-8)and cell migration(Transwell)assays,respectively,western blot(WB)was used to study the effect of knockdown of USP18 on the expression of mTOR pathway-related proteins in HepG2 cells.Results:Compared with Si-NC cells,the proliferation of Si-USP18 cells was significantly enhanced after 24,48 and 72 h of culture by CCK-8(P<0.05).Transwell assay showed the migration ability of Si-USP18 cells was significantly enhanced after 24 and 48 h as compared with Si-NC cells(P<0.05).WB assay showed that compared with Si-NC cells,the expression levels of phosphorylated mammalian target of rapamycin(p-mTOR),B-cell lymphoma-2(Bcl-2)and cyclin D1 were significantly increased in Si-USP18 cells(P<0.05).Conclusion:Downregulation of USP18 can promote the malignant behavior of hepatocarcinoma cells through mTOR signaling pathway.
