摘  要:鲎试剂广泛用于检测医药产品中的细菌内毒素,但由于鲎资源稀缺,制约了鲎试剂的生产。重组鲎C因子(rFC)作为鲎试剂的替代品,通过基因工程表达获得,可特异性检测内毒素,不依赖动物资源。综述了rFC的检测机制、发展历程及药典认可情况。研究表明,rFC方法具有特异性强、不依赖动物资源等优点,已渐被药典认可,是解决鲎试剂供应短缺的可靠方案,具有重要的应用前景。Limulus reagents are widely used to detect bacterial endotoxins in pharmaceutical products,but the production of limulus reagents is restricted due to the scarcity of limulus resources.As a substitute for limulus reagents,recombinant limulus C factor(rFC)can be obtained by gene engineering and can specifically detect endotoxins without dependence on animal resources.The study reviews the detection mechanism,development history and pharmacopoeia approval of rFC.Studies have shown that rFC method has the advantages of strong specificity and independent of animal resources,and has been gradually recognized by pharmacopoeia as a reliable solution to solve the shortage of limulus reagents.It has important application prospects.
