重复分批发酵与泡沫分离耦合生产多粘菌素E

摘  要:该文设计重复分批发酵与泡沫分离相结合的生产分离工艺,并在发酵过程中加入表面活性剂吐温80,以提高多粘菌素E的产量和下游分离工艺的效率。对重复分批发酵和吐温80添加的各项参数进行优化,并确定泡沫分离时的最佳气体体积流量。结果表明,最适的培养基置换时间为36 h,置换比例为70%,使用重复分批发酵后,每批次的发酵时间从36 h缩短至12 h,可进行6个批次的循环。吐温80在初次发酵的第24小时和置换后的第6小时添加,添加量为0.15%,气体体积流量为80 mL/min。在该优化条件下将重复分批发酵与泡沫分离耦合,在第4批次,多粘菌素E的效价达到28034.4 U/mL,高于对照组(21768.8 U/mL),消泡液中多粘菌素E效价为133706.1 U/mL,富集比和回收率分别为4.77和82.5%。This study designed a production and separation process that combines repeated batch fermentation with foam fractionation.Tween 80,a surfactant,was added during the fermentation process to enhance the yield of polymyxin E and the efficiency of the downstream separation process.Various parameters for repeated batch fermentation and the addition of Tween 80 were optimized,and the optimum gas volumetric flow rate during foam fractionation was determined.The results showed that the optimum culture medium replacement time was 36 h,with a replacement ratio of 70%.With the use of repeated batch fermentation,the fermentation time per batch was shortened from 36 h to 12 h,allowing for six cycles.Tween 80 was added at the 24th h of the initial fermentation and at the 6th h after replacement,with a concentration of 0.15%,and the gas volumetric flow rate was 80 mL/min.Under these optimized conditions,when repeated batch fermentation was coupled with foam fractionation in the 4th batch,the titer of polymyxin E reached 28034.4 U/mL,which was higher than 21768.8 U/mL in the control group.The titer of polymyxin E in the foam fraction was 133706.1 U/mL,with an enrichment ratio of 4.77 and a recovery rate of 82.5%.
