摘  要:以灵芝多糖(GLP)为分散剂,采用亚硒酸钠-抗坏血酸氧化还原法制备多糖纳米硒复合物(SeNPs),对SeNPs进行结构表征并探讨其对小鼠运动耐力的作用。结果表明:SeNPs颗粒大小67.25 nm,PDI为0.31,Zeta电位-66.72 mV;SeNPs和GLP红外光谱图高度相似,未发现新特征吸收峰,SeNPs重均分子量、数均分子量分别为7821.84 Da、6375.19 Da;SeNPs的最大热失重温度330℃,质量损失42.65%。与空白对照组相比,SeNPs低、中、高剂量组小鼠力竭游泳时间分别提高了12.69%、24.77%、37.84%;与运动模型组相比,SeNPs低、中、高剂量组小鼠血清肌酸激酶、乳酸脱氢酶活性和骨骼肌丙二醛含量均显著降低,抗氧化酶活性则显著升高,且上调了Nrf2和HO-1的蛋白表达。综上可知,SeNPs具有激活骨骼肌Nrf2信号通路,降低骨骼肌氧化应激水平的作用,进而提高小鼠运动耐力。Ganoderma lucidum polysaccharide(GLP)was used as dispersant to prepare polysaccharide nanoselenium complexes(SeNPs)by disodium selenite-ascorbic acid redox method.The structure of SeNPs was characterized and the effect of SeNPs on exercise tolerance in mice was investigated.The results showed that the particle size of SeNPs was 67.25 nm,the PDI was 0.31,and the Zeta potential was-66.72 mV.The infrared spectra of SeNPs and GLP were highly similar,and no new characteristic absorption peaks found.The weight average molecular weight and number average molecular weight of SeNPs were 7821.84 Da and 6375.19 Da,respectively.The maximum thermal weight loss temperature of SeNPs was 330℃,and the mass loss was 42.65%.Compared with the blank control group,the low,medium,and high dose groups of SeNPs increased mice exhaustion swimming time by 12.69%,24.77%,and 37.84%,respectively.Compared with the exercise model group,the activities of serum creatine kinase,lactate dehydrogenase and the content of malondialdehyde in skeletal muscle of mice in the low,middle and high dose groups of SeNPs were significantly reduced,while the activities of antioxidant enzymes were significantly increased,and the protein expressions of Nrf2 and HO-1 were up-regulated.In summary,SeNPs had the effect of activating the Nrf2 signaling pathway in skeletal muscle,reducing oxidative stress levels in skeletal muscle,and thereby improving exercise endurance in mice.
