摘 要:目的探讨BTG2在高脂饮食诱导的非酒精性脂肪肝中的表达及意义。方法将45只SPF级雄性C57BL/6小鼠随机分成为正常对照组(n=10)和模型组(n=35)。在模型组(n=35)连续高脂饮食16周,建立小鼠非酒精性脂肪肝模型。通过实时荧光定量PCR法、免疫组织化学法检测BTG2的表达量和蛋白表达水平,采用苏木精-伊红(HE)染色观察肝组织病理学改变。结果实时荧光定量PCR法检测BTG2在肝脏组织中的表达水平,与对照组相比BTG2的表达量和蛋白表达水平在模型组中显著提高。免疫组织化学法检测结果显示:与对照组相比,模型组中的BTG2蛋白质表达水平显著上调(P<0.01)。结论通过实时荧光定量PCR法、免疫组织化学法检测发现在高脂饮食诱导小鼠肝纤维化模型中的BTG2表达量均显著升高。Objective To investigate the expression and significance of BTG2 in high-fat-diet-induced NAFLD.Methods 45 male C57BL/6 mice(SPF)were randomly divided into the control group(n=10)and the model group(n=35).In the model group(n=35)with a continuous high-fat diet for 16 weeks,a mouse model of non-alcoholic fatty liver model was established.In the model group(n=35),a non-alcoholic fatty liver model was established by continuous high-fat diet for 16 weeks.The expression and protein expression levels of BTG2 were detected by qPCR and immunohistochemistry,and the histopathological changes of liver were observed by hematoxylin-eosin(HE)staining.Results Compared with the control group,the mRNA expression level and protein expression of BTG2 in the model group were significantly increased.Immunohistochemistry showed that compared with the control group,the protein expression level of BTG2 in the model group was significantly up-regulated(P<0.01).Conclusion The expression of BTG2 in high-fat diet induced mouse liver fibrosis was significantly increased by qPCR and immunohistochemistry.
