摘 要:【目的】为优化火棘干果总黄酮超声辅助提取工艺条件,掌握其抗氧化活性。【方法】在单因素试验的基础上,采用正交试验设计研究了料液比、乙醇浓度、超声功率和时间对总黄酮提取率的影响,并对总黄酮清除·DPPH自由基、ABTS+自由基、·OH自由基和NO_(2)^(-)能力进行了测定。【结果】火棘干果总黄酮的最佳提取条件为料液比1∶30 g∶mL、乙醇浓度60%、超声功率720 W、超声时间40 min,在此条件下,火棘干果总黄酮提取率为2.96%;10 mg/mL浓度下对·DPPH自由基、ABTS^(+)自由基、·OH自由基和NO_(2)^(-)清除率分别为85.26%、84%、64.8%、87%,相同浓度下清除效果略低于芦丁、山奈酚和VC标准品。【结论】研究结果为火棘干果总黄酮的提取及利用提供了参考。【Objective】This study aimed to optimize the ultrasonic extraction conditions for total flavo⁃noids from dried Pyracantha fortuneana(TFDPF)and to evaluate its antioxidant activity.【Method】Fol⁃lowing initial single-factor experiments,the effects of solid-to-liquid ratio,ethanol concentration,ultrasonic power,and ultrasonic time on the yield of TFDPF were investigated.An orthogonal experimental design was employed to refine the ultrasonic extraction parameters for TFDPF.The antioxidant capacity of TFDPF was assessed by measuring its scavenging ability against DPPH,ABTS+,hydroxyl(·OH)radicals,and NO_(2)^(-).【Result】The optimal ultrasonic extraction conditions were identified as a solid-to-liquid ratio of 1∶30 g∶mL,an ethanol concentration of 60%,an ultrasonic power of 720 W,and an ultrasonic duration of 40 minutes.Under these conditions,the TFDPF yield was approximately 2.96%.Antioxidant assays revealed that TFDPF exhibited scavenging rates of 85.26%,84%,64.8%,and 87%against DPPH,ABTS^(+),hy⁃droxyl(·OH)radicals,and NO_(2)^(-),respectively,at a concentration of 10 mg/mL.【Conclusion】The findings of this study establish a basis for the extraction and potential utilization of Pyracantha fortuneana.
