摘 要:研究了黄水的小分子化学成分,并在此基础上发掘黄水资源再利用的潜在价值。以超高效液相色谱联用质谱对黄水中的氨基酸成分进行含量测定;采用分子筛、硅胶、OSD等柱色谱及半制备高效液相色谱对黄水进行纯化分离,通过现代波谱技术对分离得到的化合物进行结构鉴定;采用细胞划痕实验探究黄水提取物对L-929细胞迁移能力的影响。结果表明:靶向测定结果显示黄水中含有19种氨基酸,其中丝氨酸、丙氨酸、酪氨酸、缬氨酸、亮氨酸含量相对最高,达到了3007.27、4883.83、3017.61、3362.24、5812.48μg/g;分离得到2个化合物,分别鉴定为11-methyl-9-hydroxyl-8,9,10,11-tetrahydro-β-carboline(1)、1,2,3,4-tetrahydro-1-methyl-β-carboline-3-carboxylic acid(2),均为氨基酸衍生物;细胞划痕结果显示:黄水提取物作用于L-929细胞24 h、36 h后划痕明显缩小,细胞迁移能力明显上升。指出了黄水中含有丰富的氨基酸成分,化合物1、2为首次从黄水中分离的氨基酸衍生物成分,且黄水提取物对L-929细胞具有明显增殖效果,说明黄水提取物具有作为营养物质的潜在作用,为黄水提供新的资源利用方向。This paper studied the small molecule chemical composition of Huangshui and explored the potential uses of Huangshui resources for reuse based on this.The content of amino acids in Huangshui are determined by using ultra-high performance liquid chromatography coupled with mass spectrometry;The Huangshui was purified by molecular sieve,silica gel,OSD column chromatography and semi-preparative high performance liquid chromatography,and the structure of the separated compounds was identified by modern spectrum technology.The effect of Huangshui extract on migration ability of L-929 cells was investigated by cell scratch test.The targeted determination results showed that Huangshui contains 19 types of amino acids,each of which is rich in content.Among them,serine,alanine,tyrosine,valine,and leucine have the highest content,reaching 3007.27,4883.83,3017.61,3362.24,5812.48,respectivelyμg/g;Two compounds were isolated and identified as 11-methyl-9-hydroxyl-8,9,10,11-tetrahydro,respectively-β-Carboline(1),1,2,3,4-tetrahydro-1-metal-β-Carboline-3carboxylic acid(2),both of which are amino acid derivatives;The cell scratch results showed that after 24 and 36 hours of treatment with Huangshui extract on L-929 cells,the scratch size significantly decreased and the cell migration ability significantly increased.It points out that Huangshui contains abundant amino acid components,and compounds 1 and 2 are the first amino acid derivative components isolated from Huangshui.Huangshui extract has a significant proliferative effect on L-929 cells,indicating that Huangshui extract has potential as a nutrient and provides new directions for resource utilization of Huangshui.
