摘  要:目的研究南极潮间带来源真菌Pseudeurotium sp.HDN17-894的次级代谢产物及其抑菌活性。方法对目标菌株进行大量发酵获取粗提物,综合利用正相、反相柱层析、半制备高效液相色谱(HPLC)以及Sephadex LH-20凝胶柱层析等分离手段对粗浸膏进行分离纯化,通过核磁共振波谱(NMR)、质谱(MS)以及单晶衍射等现代波谱学方法,同时对照相关文献数据,确定化合物的结构,并筛选得到具有抑菌活性的化合物。结果从菌株HDN17-894中分离得到1对新的吲哚-3-酮螺环外消旋体化合物(±)-pseudeurdole(1),其为首次从天然产物中发现的含吲哚-3-酮螺环己二烯酮骨架的化合物;以及2个硫赭曲菌素类化合物(2~3),3个二苯醚类化合物(4~6)和1个聚酮类化合物(7)。对化合物进行抑菌活性测试,结果显示化合物3、7对不同致病菌展现出一定的抑菌活性,其中化合物3对蜡状芽孢杆菌(Bacillus cereus)的最小抑菌浓度(MIC)为16μg/mL。Objective To study secondary metabolites from fungi Pseudeurotium sp.HDN17-894 derived from the intertidal zone of Antarctic and their antimicrobial activities.Methods The crude extract of strain HDN17-894 was obtained by large-scale fermentation.Silica gel column chromatography,semi-preparative HPLC and Sephadex LH-20 were used to separate and purify the crude extract.By comparison with data of relevant literatures,the structures of the compounds were determined by NMR,MS and single crystal diffraction.The antimicrobial compounds were screened in vitro.Results A pair of new indol-3-one spiro racemic compounds(±)-pseudeurdole(1),two known sulochrin compounds(2-3),three known diphenyl ethers(4-6)and one known polyketone compound(7)were isolated from the fungus HDN17-894.(±)-pseudeurdole(1)were compounds containing indole-3-one spirohexadienone skeleton from natural products for the first time.The antimicrobial activity of these compounds was evaluated,and the results revealed that compounds 3 and 7 showed antimicrobial potential against different pathogenic bacteria.Among them,the MIC value of compound 3 against B.cereus was 16μg/mL.
