摘  要:为研究载入虾青素对脂质体磷脂膜结构稳定性的影响,选用从鱿鱼(Sthenoteuthis oualaniensis)卵中提取的富含二十二碳六烯酸的磷脂酰胆碱(Docosahexaenoic acid-enriched phosphatidylcholine, DHA-PC)作为脂质体膜材,通过乙醇注入法制备负载虾青素的DHA-PC脂质体(Astaxanthin-embedded DHA-PC liposome, DPC-AST)和空载脂质体(Blank DHA-PC liposome, DPC),其中DHA-PC与虾青素的质量比、溶剂体积比和孵育时间是控制虾青素载入形式的关键。研究显示,当DHA-PC与虾青素的质量比为1∶0.05或1∶0.10,水与乙醇的体积比为2∶1,孵育40 min时,虾青素以J聚集体形式载入脂双层。负载J聚集体虾青素的DPC-AST的粒径(362~386 nm)明显大于DPC的粒径(57 nm)。通过多重光散射技术发现,前者具有更好的微观结构稳定性,其不稳定指数(0.5)明显小于DPC(1.2)。利用荧光偏振技术分析发现,DPC-AST的脂膜流动性较DPC显著降低,且通过改变虾青素的载入形式能够实现对脂质体膜流动性的精细调节。虾青素显示出显著的抗脂质过氧化能力,这进一步表明负载虾青素能够提高DHA-PC脂质体的膜结构稳定性。In order to investigate the effect of embedded astaxanthin on the stability of phospholipid membranes structure of liposomes,docosahexaenoic acid-enriched phosphatidylcholine(DHA-PC)extracted and purified from squid roe was selected as the liposome membrane material to prepare astaxanthin-embedded DHA-PC liposome(DPC-AST)and blank DHA-PC liposome(DPC)with ethanol injection method.It was found that the mass ratio of DHA-PC to astaxanthin,as well as solvent volume ratio and incubation time were key factors to control the form of embedded astaxanthin.Astaxanthin J-aggregates can be successfully embedded in lipid bilayers when the mass ratio of DHA-PC to astaxanthin was 1∶0.05 or 1∶0.10,the volume ratio of water to ethanol was 2∶1,and incubation time was 40 min.The particle size(362~386 nm)of DPC-AST embedded with J-aggregates astaxanthin was significantly larger than that of DPC(57 nm).The instability index(0.5)of DPC-AST embedded with J-aggregates astaxanthin was significantly lower than that of DPC(1.2)as determined by multiple light scattering technologies.DPC-AST embedded with J-aggregates astaxanthin had better microstructure stability.The fluorescence polarization analysis showed that the membrane fluidity of DPC-AST is decreased in comparison with that of DPC.Meanwhile,the fine regulation of liposome membrane fluidity could be achieved by changing the embedding form of astaxanthin.Moreover,the improved anti-lipid peroxidation capacity of astaxanthin further proved that astaxanthin can significantly improve the stability of the membrane structure of DHA-PC liposomes.
