摘  要:采用单因素实验与响应面法优化地稔多糖超声-复合酶法提取工艺,运用高效液相色谱法和红外光谱法分析地稔多糖组成与结构特征,通过检测地稔多糖对2,2-联氨-双-3-乙基苯并噻唑啉-6-磺酸(2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基、1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基和羟基自由基的消除能力探究其抗氧化能力。结果表明,在酶解条件为木瓜蛋白酶比例1.9%、纤维素酶比例3.0%、酶解温度67℃、酶解时间70 min时,地稔多糖提取率达到最高为(1.364±0.004)%。地稔多糖由8种单糖组成,其中半乳糖醛酸、半乳糖和阿拉伯糖为主要成分。地稔多糖具有ABTS阳离子自由基消除能力(IC_(50)=3.047 mg/mL)、DPPH自由基消除能力(IC_(50)=26.93μg/mL)及羟基自由基消除能力(IC_(50)=7.457 mg/mL),具有较强的抗氧化能力,且抗氧化强度呈现剂量依赖性。研究结果可为地稔多糖的开发和应用提供了理论依据。Single factor test and response surface method were used to optimize the ultrasound-complex enzymatic extraction process of Melastoma dodecandrum Lour.polysaccharide(MDP).The composition and structural characteristics of MDP were analyzed by high performance liquid chromatography and infrared spectroscopy,and the antioxidant capacity of MDP was explored by detecting its ability to eliminate 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation and hydroxyl radicals.The results showed that under the conditions of 1.9%papain,3.0%cellulase,67℃,and 70 min,the yield of MDP reached the highest((1.364±0.004)%).Eight monosaccharide components were identified in the polysaccharide,of which galacturonic acid,galactose and arabinose were the main components.MDP exhibited ABTS radical cation eliminating ability(IC_(50)=3.047 mg/mL),DPPH radical eliminating ability(IC_(50)=26.93μg/mL)and hydroxyl free radical eliminating ability(IC_(50)=7.457 mg/mL),and had strong antioxidant ability.The antioxidant strength showed a dose-dependent manner.This study provides a theoretical basis for the development and application of MDP.
