摘  要:采用色谱柱SB-C 18(250 mm×4.6 mm,5μm);流动相为:三氯甲烷∶乙腈∶甲醇=3∶12∶85,含抗坏血酸0.4 g/L;流速:2 mL/min等度洗脱;检测波长:450 nm,建立一个β-胡萝卜素的高效液相色谱测定方法。凝胶糖果样品取完整多颗破碎混匀后,在避光且添加抗氧化剂情况下分散、提取、皂化、液液萃取、水洗、浓缩后复溶测试,在β-胡萝卜素含量0.25~10 mg/L范围内线性系数R=1.00000,呈良好的线性;各前处理影响因子经摸索后设置为最佳,此时重复性变异系数为0.32%,中间精密度相对标准偏差为1.45%,回收率为95.37%~99.93%。该方法体现出良好的准确度、精密度、耐用性,适用于凝胶糖果中β-胡萝卜素的测试。The determination was performed on SB-C 18(250 mm×4.6 mm,5μm)with the mobile phase consisted of trichloromethane∶acetonitrile∶methanol=3∶12∶85,containing ascorbic acid 0.4 g/L,at the flow rate of 2 mL/min.The detection wavelength was 450 nm.A HPLC method for the determination ofβ-carotene was established.After the gel candy samples were broken and mixed,the specimen was dispersed,extracted,saponified,liquid liquid extraction,water washed,concentrated and redissolved in sequence,under the condition of avoiding light and adding antioxidants.The linear coefficient R was 1.00000 in the range ofβ-carotene content 0.25~10 mg/L,showing a good linearity.The influence factors of pretreatment were set as the best,the coefficient of variation of repeatability was 0.32%,the RSD of intermediate precision was 1.45%,and the recovery was 95.37%~99.93%.This method showed good accuracy,precision and durability,and was suitable for the determination ofβ-carotene in gel candy.
