摘  要:优化婴幼儿配方食品中维生素A和维生素E含量检测的皂化-RPHPLC法和酶解-NPHPLC法,并对检验结果进行对比研究。结果表明:将婴幼儿配方食品样本复溶后再检测其中的维生素A和维生素E含量,提高了检测结果代表性,有利于批量化检测;乙酸乙酯-正己烷溶液(3∶2,v/v)为提取剂时,提取效果好且环保。维生素A和维生素E含量的检测结果分别在各自对应的浓度范围内呈良好线性关系,相关系数R2≥0.999,皂化-RPHPLC法中维生素A和维生素E定量限在27.3~142μg/100g之间,酶解-NPHPLC法中维生素A和维生素E定量限在10.8~112μg/100g之间,酶解-NPHPLC法灵敏度更高。在低中高三个添加水平下,维生素A加标回收率在82.2%~96.8%之间,RSD在1.0%~4.6%之间;维生素E加标回收率在83.5%~100.6%之间,RSD在1.9%~9.7%之间。皂化-RPHPLC法和酶解-NPHPLC法检验结果无显著性差异。优化后的检测方法简单高效,样品代表性好。The saponification-RPHPLC and enzymatic hydrolysis-NPHPLC methods for the determination of vitamin A and vitamin E contents in infant formula were optimized and the detection results were compared and analyzed.The results showed that the redissolved sample detection improved the representativeness of the detection results and was conducive to the batch detection.When ethyl acetate-n-hexane solution(3∶2,v/v)was used as the extraction agent,the extraction effect was good and environmentally friendly.The detection results of vitamin A and vitamin E contents showed good linear relationships within their respective concentration ranges,and the correlation coefficient R2≥0.999.The quantitation limits of vitamin A and vitamin E in the saponification-RPHPLC method ranged from 27.3 to 142μg/100g.The quantitation limits of vitamin A and vitamin E in enzymatic hydrolysis�NPHPLC method were 10.8~112μg/100g,and the sensitivity of enzymatic hydrolysis-NPHPLC method was higher.At three levels of low,medium,and high additions,the spike recovery rates of vitamin A were between 82.2%and 96.8%,and the RSD was between 1.0%and 4.6%.The spike recovery rates of vitamin E were between 83.5%and 100.6%,and RSD was 1.9%~9.7%.There was no significant difference between saponification-RPHPLC and enzymatic hydrolysis-NPHPLC.The optimized detection method was simple,efficient,and ensured good sample representativeness.
