摘 要:目的基于定向固定化抗体的侧向流免疫检测试纸建立食用油中玉米赤霉烯酮(zearalenone,ZEN)的检测方法,并考察市售食用油中ZEN污染状况。方法通过链球菌蛋白G将ZEN抗体定向固定化在铕荧光微球上制备了免疫探针,构建侧向流免疫检测(lateral flow immunoassay,LFIA)试纸,并建立了食用油中ZEN含量的快速检测方法,并对65个市售食用油样品中的ZEN含量进行了检测。结果采用抗体定向固定化方式制备免疫探针可以使检测曲线线性范围收窄,提高试纸的检测精密度。建立的食用油中ZEN快速检测方法前处理步骤只需1步,检测用时仅15 min。方法的检出限为3.93μg/kg,线性范围为22.40~800.00μg/kg。市售食用油样品中ZEN的检出率为58.5%,其中,玉米油中的ZEN检出率为100%,且ZEN含量最高。结论本研究成功制备了可以快速、准确检测食用油中ZEN含量的侧向流免疫层析试纸,并且所探究的抗体定向固定化技术对于提高其他小分子污染物免疫检测技术的分析性能具有参考价值。Objective To establish a method for the detection of zearalenone(ZEN)in cooking oil based on the lateral flow immunoassay strip with directional immobilized antibodies,and to investigate the contamination status of ZEN in commercial cooking oil.Methods The immunoprobes were generated through the oriented immobilization of ZEN antibodies on europium nano particles with the aid of Streptococcus protein G.These immunoprobes were then utilized in the construction of lateral flow immunoassay(LFIA)test strips.This approach enabled the rapid detection of ZEN levels in edible oil.Subsequently,the ZEN content in 65 commercially available edible oil samples was assessed using this novel detection method.Results Utilizing antibody-oriented immobilization to create immunoprobes could refine the linear range of the detection curve and enhance the accuracy of the test strip.The established rapid ZEN detection method for edible oils involves only a single pretreatment step,with a detection time of just 15 minutes,highlighting its ease and efficiency.The method had a detection limit of 3.93μg/kg,a linear range from 22.40 to 800.00μg/kg.The detection rate of ZEN in commercial edible oil samples was 58.5%,with corn oil exhibiting a 100%detection rate and the highest ZEN content detected.Conclusion This study successfully developed a LFIA test strip for rapid and accurate ZEN content detection in edible oils,demonstrating its applicability to real sample analysis.Furthermore,the oriented immobilization technology for antibody investigated in this study offers insights for enhancing the analytical performance of other small molecule pollutant immunodetection techniques.
