摘 要:目的:探讨制备羚羊角多肽的最佳酶解工艺条件。方法:以羚羊角多肽提取率为指标筛选中性蛋白酶、胃蛋白酶、胰蛋白酶中酶解羚羊角多肽作用较佳的蛋白酶。采用单因素法初步考察胰蛋白酶用量、酶解温度、pH、酶解时间等因素对多肽提取率的影响;再以响应面法考察酶用量、酶解时间、酶解温度对羚羊角多肽提取率的影响,确定最佳酶解工艺﹐并进行验证。结果:响应面得到最佳提取工艺参数为:提取温度55℃,提取时间3.4 h,胰蛋白酶用量5400 U/mL。结论:响应面法优化的羚羊角多肽提取工艺简单快捷、稳定,成本低廉,安全性高,为羚羊角的进一步开发利用提供参考。Objective:To study the optimum enzymatic hydrolysis process for the extraction of antelope horn polypeptide.Method:Using the extraction rate of polypeptide as an indicator,enzyme with better extraction effects were screened from neutral protease,pepsin and trypsin.The effects of trypsin dosage,enzymolysis temperature,pH,enzymolysis time and other factors on the extraction rate of polypeptide were investigated by single factor method.Then the effects of enzyme dosage,enzymatic hydrolysis temperature and enzymatic hydrolysis temperature on the extraction rate of antelope horn polypeptide were investigated by response surface method,and the optimal enzymatic hydrolysis process was determined and verified.Result:The optimal hydrolysis process was as follows:reaction temperature of 55℃,extraction time of 3.4 h,trypsin dosage of 5500 U/mL.Conclusion:The enzymatic hydrolysis process of antelope horn polypeptide optimized by response surface method is simple,fast,stable,low cost and high safety,which provides a reference for further development and utilization of antelope horn.
