摘 要:为了提高刺梨果渣的利用率,采用天然低共熔溶剂配合超声辅助法提取刺梨果渣中的有效成分,包括没食子酸、芦丁、原花青素b2、异槲皮苷、儿茶素。制备了10种以氯化胆碱为氢键受体的低共熔溶剂,采用高效液相色谱(HPLC)分析法对提取过程中的各个参数进行单因素优化,并进行响应面分析,之后使用分光光度法测定刺梨果渣提取物的抗氧化活性。结果表明:由氯化胆碱和DL-乳酸组成的天然低共熔溶剂配合超声辅助对于刺梨果渣中5种成分具有更高的提取效率;进一步研究该低共熔溶剂的氢键受体与供体物质的量比、溶剂含水率、m(料)∶V(液)、提取温度、提取时间对刺梨果渣中5种有效成分的提取率的影响,并采用响应面法优化m(料)∶V(液)、提取时间和提取温度,得出结果,氯化胆碱与DL-乳酸物质的量比为1∶3、溶剂含水率为20%、m(料)∶V(液)为1 g∶21 mL、提取温度为60℃、提取时间为38 min时,为最佳提取工艺,刺梨果渣中没食子酸、芦丁、原花青素b2、异槲皮苷、儿茶素的提取率分别为64.45、1036.55、4592.59、5179.53、2308.35μg/g。将优化后的提取工艺与传统提取工艺进行对比,证明了该提取方法的优越性。此外,该提取方法得到的提取物DPPH自由基清除半抑制质量浓度为0.182 g/L,抗氧化效果较为显著。To enhance the utilization of Rosa roxburghii pomace,a natural deep eutectic solvent combined with ultrasound-assisted extraction was employed to extract the active components from R.roxburghii pomace.The target compounds included gallic acid,rutin,proanthocyanidin b2,isoquercitrin,and catechin.Ten types of natural deep eutectic solvent using choline chloride as the hydrogen bond acceptor were prepared.High performance liquid chromatography(HPLC)was used to single-factor optimise the parameters of the extraction process,and response surface analysis was performed.The antioxidant activity of the R.roxburghii pomace extract was then determined using the spectrophotometric method.The results showed that the natural deep eutectic solvent composed of choline chloride and DL-lactic acid combined with ultrasound-assisted extraction provided higher extraction efficiency for the five components from R.roxburghii pomace.Further studies investigated the effects of the molar ratio of the hydrogen bond acceptor to donor,water content of the solvent,solid-to-liquid ratio,extraction temperature,and extraction time on the extraction yields of the five active components from R.roxburghii pomace.Response surface methodology was used to optimize the solid-to-liquid ratio,extraction time,and extraction temperature.The optimal extraction conditions were determined as follows:a molar ratio of choline chloride to DL-lactic acid of 1∶3,a solvent water content of 20%,a solid-to-liquid ratio of 1 g∶21 mL,an extraction temperature of 60℃,and an extraction time of 38 minutes.Under these conditions,the extraction yields of gallic acid,rutin,proanthocyanidin b2,isoquercitrin,and catechin were 64.45,1036.55,4592.59,5179.53,and 2308.35μg/g,respectively.Comparing the optimized extraction process with traditional methods demonstrated the superiority of this approach.In addition,the extract obtained by this extraction method had a DPPH radical scavenging semi-inhibitory mass concentration of 0.182 g/L,which showed a more significant antioxi
