摘 要:目的:制备铁皮石斛叶多糖组分,探究多糖提取物基本组成、理化性质与体外抗氧化活性,为铁皮石斛叶资源开发利用提供依据。方法:以铁皮石斛叶为原料,通过水提醇沉工艺结合大孔树脂脱色脱蛋白制备出铁皮石斛叶多糖。通过理化检测分析其化学组成,利用色谱法测试其分子质量与单糖组成,并初步探讨其流变学性质与体外抗氧化活性。结果:经组成分析发现,该多糖总糖含量为49.40%、糖醛酸含量为9.60%、蛋白含量0.18%、水分含量12.70%、灰分含量20.70%,所得铁皮石斛叶多糖为高灰分、低糖醛酸的多糖;单糖组成研究表明该多糖主要由甘露糖、葡萄糖、半乳糖组成,同时含有少量阿拉伯糖及半乳糖醛酸,摩尔比为甘露糖:葡萄糖:半乳糖:阿拉伯糖:半乳糖醛酸=0.597:0.176:0.149:0.028:0.049;流变学研究表明铁皮石斛叶多糖溶液属于非牛顿流体,呈现出剪切稀化现象;抗氧化测试结果表明铁皮石斛叶多糖具有潜在的抗氧化能力,特别是对羟基自由基具有良好的清除能力,当浓度为2 mg/mL时,清除率达81.8%。结论:铁皮石斛叶多糖资源丰富,具有良好的抗氧化活性,值得深入研究开发。Objective:To provide experiment basis for the development and utilization of Dendrobium officinale leaf resources.Polysaccharides from leaf of Dendrobium officinale Kimura et Migo (LDOP) were prepared.Furthermore,the basic composition,physicochemical properties,and antioxidant activity in vitro of polysaccharide extracts were preliminarily explored.Method:Using Dendrobium officinale Kimura et Migo leaves as raw materials,polysaccharides were prepared by water extraction and alcohol precipitation process combined with macro-porous resin for decolorization and deproteinization.The chemical composition was analyzed,and its molecular weight and monosaccharide composition were tested by chromatography.The rheological properties and antioxidant activity were preliminarily explored in vitro.Result:According to the composition analysis,it was found that the total sugar content of LDOP was 49.40%,the amount of uronic acid,protein,moisture,and ash were 9.60%,0.18%,12.70%,20.70%,respectively.And the results showed that the LDOP were the polysaccharides with high ash and low uronic acid;Monosaccharide composition study showed that the polysaccharide was mainly composed of mannose,glucose,and galactose,with a small amount of arabinose and galacturonic acid.The molar ratio was:mannose:glucose:galactose:arabinose:galacturonic acid=0.597:0.176:0.149:0.028:0.049;Rheological studies had shown that the solution of LDOP belong to a non-Newtonian fluid,and exhibit shear thinning phenomenon;The antioxidant test results indicated that the LDOP had potential antioxidant capacity,especially good scavenging ability against hydroxyl radicals.When the concentration was 2 mg/mL,the scavenging rate was up to 81.8%.Conclusion:The polysaccharide resources of Dendrobium officinale Kimura et Migo leaves are abundant and have good antioxidant activity,which is worth further research and development.
