摘 要:利用一种更加高效、准确的双波长比色法测定高粱支链淀粉占总淀粉的比例,相较于传统的单波长比色法或双波长比色法,缩短样品前处理的时间,减少间接试验的步骤。试验通过将直链淀粉标准溶液和支链淀粉标准溶液在400~900 nm波长范围内全扫描后,选用535和630 nm这2个吸光度较大波长测量高粱中支链淀粉的比例,验证其回收率(R),平均回收率为98.64%,回收率相对标准偏差为2.17%,验证测量重复性,标准偏差(S)为0.16%,变异系数(C.V.)为0.16%,检出限(S_(LOD))为4.15%,定量限(S_(LOQ))为8.70%。该方法测定高粱支链淀粉比例快捷准确,能为快速判断高粱品质提供依据。A more efficient and accurate dual-wavelength colorimetric method was used to determine the ratio of amylopectin to total starch in sorghum.Compared with the traditional single-wavelength colorimetric method or dual-wavelength colorimetric method,the pretreatment time of samples was shortened and the indirect test steps were reduced.In the experiment,amylopectin standard solution and amylopectin standard solution were fully scanned in the wavelength range of 400-900 nm,and 535 nm and 630 nm wavelength with large absorbance were selected to measure the proportion of amylopectin in sorghum.The recovery rate(R)was verified.The average recovery rate was 98.64%,and the relative standard deviation of recovery rate was 2.17%.The standard deviation(S)was 0.16%,the coeficient of variation(C.V.)was 0.16%,the detection limit(SLoD)was 4.15%,and the limit of quantification(SLoo)was 8.70%.This method was quick and accurate to determine the proportion of amylopectin in sorghum,and could provide a basis for rapid judgment of sorghum quality.
