摘 要:为优化桃胶多糖的提取工艺并研究其体外免疫作用,采用加压碱水法提取桃胶多糖,在单因素试验结果的基础上,通过响应面试验对其提取工艺进行优化。结果表明,桃胶多糖最佳提取工艺条件为:液料比30∶1(mL/g),提取压力1.0 MPa,提取时间35 min,在此条件下,桃胶多糖得率为85.59%。体外免疫作用试验结果表明:桃胶多糖能够增加小鼠的体重和脏器指数,增加小鼠的细胞因子肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)、γ-干扰素(Interferon-γ,IFN-γ)和白细胞介素-6(Tnterleukin-6,IL-6)含量,增加小鼠的免疫球蛋白A(IgA)、免疫球蛋白G(IgG)和免疫球蛋白M(IgM)含量,表明桃胶多糖具有提高免疫力的作用。该研究为桃胶多糖的高效提取及开发利用提供了参考。To optimize the extraction process of peach gum polysaccharide(PGP)and investigate its immune effects in vitro,the method of extracting peach gum polysaccharide was used by pressurized alkaline water.Based on singlefactor analysis,the extraction process was optimized using response surface methodology.The results showed that the optimal extraction process conditions were liquid-solid ratio of 30∶1(mL/g),extraction pressure of 1.0 MPa,and extraction time of 35 minutes.Under the optimal extraction conditions,the PGP yield was 85.59%.The results of immune effect experiments in vitro showed that peach gum polysaccharide could increase the body weight and organ index of mice,increase the contents of tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)and tnterleukin-6(IL-6)in mice cytokine,and increase the levels of immunoglobulin A(IgA),immunoglobulin G(IgG),and immunoglobulin M(IgM)in mice,which indicating that peach gum polysaccharide had the effect of enhancing immunity.This study provides a reference for the efficient extraction,development and utilization of peach gum polysaccha rides.
