摘 要:为解决聚乙烯(PE)包装材料在土壤中难以降解的问题,从土壤中分离出能够降解PE保鲜膜的降解菌。经逐步筛选纯化,对菌株D-1进行形态学观察、生理生化鉴定和16S rDNA序列分析;将菌株D-1接种到以PE膜片为唯一碳源的无机盐液体培养基中,在35℃,150 r/min培养。试验结果表明,菌株D-1为白色嗜盐多孢放线菌,28 d后PE膜失重率为3.34%;电镜观察发现PE膜表面有明显孔洞和裂痕;红外扫描显示PE膜中引入了碳碳三键、羧基、醚键等活性官能团,漆酶活力为8.7 U/L。研究为PE膜的降解提供科学依据,并丰富了降解菌株的资源库。To solve the problem of difficult degradation of polyethylene(PE)packaging film,a strain from soil that can degrade PE plastic was isolated.After screening and purification,morphological observation,physiological and biochemical identification,and 16S rDNA sequence cloning were done to identify the strain D-1.Then the strain was inoculated on simple inorganic salt medium with PE film as the sole carbon source and cultured at 35℃,150 r/min.The results show that the strain D-1 is identified as Haloactinopolyspora alba.After 28 d incubation,the weight loss of PE film reaches 3.34%.Many holes and fissure were observed on the surface of PE film by SEM;and oxygen-containing functional groups such as carbon-carbon triple bond,ether bond and carboxyl group were introduced into the PE film through infrared scanning,and the laccase activity was 8.7 U/L.This study provides a scientific basis for the degradation of PE film,and enriches the resource bank of the degrading strains.
