摘 要:乳铁蛋白是人体液中含量丰富的糖蛋白,是转铁蛋白家族的成员之一,具有广泛的生理和营养功能,准确定量乳铁蛋白的含量具有重要的意义。然而,由于目前还缺少乳铁蛋白的标准物质,导致复杂基体中乳铁蛋白的测量结果偏差较大,且无法溯源。本文对乳铁蛋白纯物质的测量及标准物质的定值方法进行了介绍,包括氨基酸同位素稀释质谱法和质量平衡法。在此基础上,对人乳和奶粉等乳品复杂基质中乳铁蛋白的检测技术进行了介绍,包括乳及乳制品中的乳铁蛋白常用的高效液相色谱法、免疫测定法、高效液相色谱-串联质谱法,对基于荧光的适配体传感器、电化学传感器和表面等离子体共振传感器方法的原理进行了简要的分析,最后对几种分析方法之间的分析性能进行了比较,对于推动乳铁蛋白测量分析技术的进一步研究具有指导意义。Lactoferrin is a rich glycoprotein in human fluids and a member of the transferrin family.Lactoferrin has a wide range of physiological and nutritional functions,and accurate quantification of lactoferrin content is of great significance.However,due to the current lack of standard substances for lactoferrin,the measurement results of lactoferrin in complex matrices have significant deviations and cannot be traced.This paper introduced the measurement of pure lactoferrin substances and the calibration methods of standard substances,including amino acid isotope dilution mass spectrometry and mass balance method.On this basis,this paper introduced the detection techniques for lactoferrin in complex matrices such as human milk and milk powder,including commonly used high performance liquid chromatography,enzyme-linked immunosorbent assay,and high performance liquid chromatography-tandem mass spectrometry for lactoferrin in milk and dairy products,as well as fluorescence based aptamer sensors,electrochemical sensors,and surface plasmon resonance sensors.Finally,compared the analytical performance of several analysis methods,which has guiding significance for further research on lactoferrin measurement and analysis technology.
