摘 要:目的研究花生受黄曲霉菌侵染后代谢物及关键代谢途径的变化,以了解花生籽仁应答黄曲霉菌侵染的生理生化机制。方法基于非靶向代谢组学技术,结合多元统计分析方法,分析不同花生品种应答黄曲霉菌侵染时差异代谢物及代谢途径变化。结果共发现了349个差异代谢物,其中中花6号品种中有268个,远杂9102品种中有238个,这些差异代谢物主要包括类黄酮类、萜类化合物、苯及其衍生物、聚酮类、脂肪酰基、氨基酸类和苯丙烷类。对差异代谢物进行代谢途径分析表明,中花6号中不饱和脂肪酸生物合成、苯丙氨酸代谢、氨酰基tRNA的生物合成、α-亚麻酸代谢通路对黄曲霉菌的响应速度更快。结论苯丙氨酸代谢中的白藜芦醇、肉桂酸、香豆酸和阿魏酸以及亚麻酸代谢途径中的13S-HPODE在响应黄曲霉菌侵染中起重要作用,为早期发现黄曲霉菌感染和选育抗黄曲霉毒素污染的花生品种提供指导。Objective To study the changes of metabolites and key metabolic pathways in peanut infected by Aspergillus flavus,and understand the physiological and biochemical mechanism of peanut kernel response to Aspergillus flavus infection.Methods Non-targeted metabolomics and multivariate statistical analysis were used to analyze the changes of metabolites and metabolic pathways in different peanut cultivars in response to Aspergillus flavus infection.Results A total of 349 metabolites were identified,including 268 in Zhonghua 6 and 238 in Yuanza 9102,these differential metabolites mainly included flavonoids,terpenoids,benzene and its derivatives,polyketones,fatty acyl groups,amino acids and phenylpropanes.Metabolic pathway analysis of differential metabolites showed that the biosynthesis of unsaturated fatty acids,phenylalanine metabolism,aminoacyl tRNA biosynthesis andα-linolenic acid metabolic pathway in Zhonghua 6 were more responsive to Aspergillus flavus.Conclusion Resveratrol,cinnamic acid,coumaric acid,ferulic acid in the phenylpropanoidand metabolism and13S-HPODE in the linolenic acid pathway play important roles in the response to Aspergillus flavus infection,to provide guidance for early detection of Aspergillus flavus infection and selection of peanut varieties resistant to aflatoxins contamination.
