摘  要:目的 探讨核基质结合区结合蛋白1 (SATB1)和表皮生长因子受体(EGFR)在胃癌中功能及其临床意义。方法 收集石蜡包埋的胃癌病理切片60例和胃黏膜慢性炎症组织切片39例,采用免疫组化方法检测SATB1和EGFR的表达情况。采用干扰小RNA(siRNA)瞬时转染入胃癌细胞,实时荧光定量反转录聚合酶链反应(RT-qPCR)和Western blot检测转染后SATB1和EGFR的表达,再通过细胞划痕及Transwell实验检测转染后胃癌细胞MGC803的迁移及侵袭变化。KM生存分析不同SATB1和EGFR表达胃癌患者的预后情况。结果 胃癌组织中SATB1和EGFR的表达均高于胃黏膜慢性炎症组织,差异有统计学意义(P<0.05)。SATB1的表达与胃癌淋巴结转移以及远处转移相关,差异有统计学意义(P<0.05);EGFR的表达也与远处转移相关,差异有统计学意义(P<0.05)。SATB1和EGFR在胃癌组织中的表达有一定的相关性(r=0.49,P<0.05)。RT-qPCR和Western blot实验表明siRNA能有效敲低胃癌细胞中SATB1和EGFR的表达;细胞划痕及Transwell实验结果显示,敲低SATB1和EGFR可致使胃癌细胞的迁移和侵袭受到抑制。KM生存曲线结果表明SATB1低表达的胃癌患者预后相对较好,EGFR低表达的胃癌患者预后好。结论 SATB1和EGFR的表达水平与胃癌的淋巴结转移及远处转移密切相关,且降低SATB1和EGFR的表达能抑制胃癌的迁移和侵袭。Objective To investigate the function and clinical significance of nuclear matrix binding region binding protein 1(SATB1)and epidermal growth factor receptor(EGFR)in gastric cancer.Methods The expression of SATB1 and EGFR was detected by immunohistochemistry in 60 cases of paraffin-embedded gastric cancer and 39 cases of gastric mucosal inflammation.Small interfering RNA(siRNA)was transfected into gastric cancer cells.The expression of SATB1 and EGFR after transfection was detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-qPCR)and Western blot.The cell migration and invasion changes of MGC803 after transfection were detected by cell scratch test and Transwell test.The prognosis of gastric cancer patients with different SATB1 and EGFR expression was analyzed by KM survival analysis.Results The expression of SATB1 and EGFR in gastric cancer tissues was higher than that in gastric mucosa inflammation tissues,and the difference was statistically significant(P<0.05).The expression of SATB1 was correlated with lymph node metastasis and distant metastasis of gastric cancer,and the difference was statistically significant(P<0.05).EGFR expression was also associated with distant metastasis,and the difference was statistically significant(P<0.05).There was a positive correlation between the expression of SATB1 and EGFR in gastric cancer tissues(r=0.49,P<0.05).RT-qPCR and Western blot showed that siRNA could effectively knock down the expression of SATB1 and EGFR in gastric cancer cells.The results of cell scratch and Transwell experiments showed that the migration and invasion of gastric cancer cells were inhibited by knocking down SATB1 and EGFR.The results of KM survival curve showed that gastric cancer patients with low expression of SATB1 had a better prognosis,while those with low expression of EGFR had a better prognosis.Conclusion The expression levels of SATB1 and EGFR are closely related to lymph node metastasis and distant metastasis of gastric cancer,and decreasin
